Circulating miRNAs in acute new-onset atrial fibrillation and their target mRNA network

BackgroundMicroRNAs (miRNAs) are involved in the pathogenesis of atrial fibrillation (AF), acting on development and progression. Our pilot study investigated the expression of six miRNAs and their miRNA-mRNA interactions in patients with acute new-onset AF, well-controlled AF, and normal sinus rhythm (controls). Methods and resultsPlasma of acute new-onset AF patients (n=5) was collected in the emergency room when patients presented with irregular and fast-atrial fibrillation rhythm. Samples from well-controlled AF (n=16) and control (n= 15) patients were collected during medical appointments following an ECG. Expression of miR-21, miR-133a, miR-133b, miR-150, miR-328, and miR-499 was analyzed by real-time PCR. Ingenuity Pathway Analysis and the TargetScan database identified the top 30 mRNA targets of these miRNA, seeking the miRNA-mRNA interactions in cardiovascular process. Increased expression of miR-133b (1.4-fold), miR-328 (2.0-fold), and miR-499 (2.3-fold) was observed in patients with acute new-onset AF, compared with well-controlled AF and control patients. Decreased expression of miR-21 was seen in patients with well-controlled AF compared to those with acute new-onset AF and controls (0.6-fold). The miRNA-mRNA interaction demonstrated that SMAD7 and FASLG genes were the targets of miR-21, miR-133b, and miR-499 and were directly related to AF, being involved in apoptosis and fibrosis. ConclusionThe miRNAs had different expression profiles dependent on the AF condition, with higher expression in the acute new-onset AF than well-controlled AF. Clinically, this may contribute to an effective assessment for patients, leading to early detection of AF and monitoring to reduce the risk of other serious cardiovascular events.

Summary: Familial hypercholesterolemia (FH) is caused by harmful mutations in the LDLR gene, resulting in significantly increased low-density lipoprotein (LDL) cholesterol and premature atherosclerotic cardiovascular disease. In this study, the functional effects of LDLR variants in Brazilian FH patients were assessed. It was found that LDLR null and defective variants reduce LDL binding and uptake in activated T-lymphocytes, while LDLR missense variants affect LDLR conformational stability and the dissociation of the LDLR-ApoB complex.

Long non-coding RNA and circular RNA: new perspectives for molecular pathophysiology of atrial fibrillation

Summary: Many studies have shown that atrial fibrillation (AF) is associated with endogenous genetic regulatory mechanisms, particularly long non-coding RNAs (lncRNAs) and circular RNAs (circRNAs), which play a role in AF development and related diseases. This review examines studies that have analyzed the expression of lncRNAs and circRNAs in AF, revealing their importance in the pathophysiological process and suggesting their involvement in miRNA or mRNA regulation. The lack of overlap among the studies may be attributed to variations in sample types, methods, species, and patient classification.

Dynamics of the personalities of PCSK9 on missense variants (rs505151 and rs562556) from elderly cohort studies in Brazil

Summary: The Proprotein Convertase Subtilisin/Kexin Type 9 (PCSK9) plays a role in lipid metabolism and can lead to coronary artery disease. Large-scale genomic studies have been conducted worldwide to support precision medicine, but non-European populations are underrepresented in public genomic data banks. However, two high-frequency variants were found in the ABraOM databank from a study conducted in Sao Paulo, Brazil. Molecular dynamics study revealed the structural and dynamical features of these variants and highlight the importance of the prodomain in PCSK9 dynamics.

Summary: In order to prevent biofouling and combat the COVID-19 virus, PVC nanocomposites with silver nanoparticles (AgNPs) were prepared. The PVC/AgNP nanocomposites exhibit virucidal activity against SARS-CoV-2 when the AgNP content is at least 0.3 wt%, making them suitable for manufacturing furniture and medical equipment with self-disinfectant capacity to prevent secondary routes of COVID-19 transmission.

Summary: This study investigated the individual and collaborative role of BRAF, TERT, HLA-G, and microRNAs in papillary thyroid carcinoma (PTC). It was found that HLA-G and BRAF were strongly expressed in PTC specimens, and BRAF overexpression was associated with poor response to therapy. BRAF(V600E) and TERTC228T mutations were associated with extrathyroidal extension, advanced age, and advanced-stage cancer. Nine modulated microRNAs targeting BRAF, TERT, and/or HLA-G genes were observed in PTC, indicating their involvement in cancer-related signaling pathways.
