Ecto-5'-nucleotidase/CD73 contributes to the radiosensitivity of T24 human bladder cancer cell line

Trimodal therapy is a reasonable bladder-preserving option to radical cystectomy. However, many tumors are radioresistive. In this sense, the identification of new prognostic and predictive biomarkers that allow the selection of patients with better responses to radiation therapy would improve outcomes. With the aim of using ecto-5'-nucleotidase/CD73 as a predictive biomarker, the role of this enzyme in the context of radiotherapy in T24 human bladder cancer cell line was investigated. T24 cell line was exposure to a single dose of radiation (4 Gray) and trypan blue assay (pharmacological assays of viability/cumulative population doubling), flow cytometry (cell cycle/cell death/active caspase-3/ecto-5'-nucleotidase/CD73 protein staining), DAPI staining (nuclear morphometric assay), RT-PCR and real-time PCR, malachite green method (ectonucleotidase enzymatic assay), and HPLC (analysis of AMP metabolism) were carried out. T24 cell line in which ecto-5'-nucleotidase/CD73 has been completely silenced (5'KO) was also used. The exposure of T24 cell line to a single dose (4 Gray) of radiation-induced cell death and triggered a transitory increase in ecto-5'-nucleotidase/CD73 expression, increased ectonucleotidase activity, and led to adenosine and inosine accumulation in the extracellular medium. Pharmacological inhibition or knocking out ecto-5'-nucleotidase/CD73 rescued cells' proliferative capacity, reducing their sensitivity to radiation. Our findings show that the induction of ecto-5'-nucleotidase/CD73 by radiation contributes to the radiosensitivity of T24 cell line.

Summary: This study compared three non-sequencing protocols for detecting major NPM1 mutations and evaluated nuclear morphometric analysis as an alternative method for cuplike blast detection. The results showed that quantitative PCR (qPCR) is the best option to identify NPM1 mutations, with ARMS-PCR being a cheaper alternative. Multiparameter flow cytometry (MFC) can be used as a screening method.

Summary: A series of adamantyl carboxamide derivatives containing sulfonate or sulfonamide moiety were designed and evaluated for their antiproliferative activity against NCI-60 cancer cell lines panel. Compounds 1e and 1i exhibited potent and broad-spectrum inhibition against multiple cancer subtypes while showing safety towards normal cells.

Targeting ecto-5′-nucleotidase: A comprehensive review into small molecule inhibitors and expression modulators

Summary: Purinergic signaling is being explored as a potential therapeutic approach for cancer treatment. ATP acts as a chemotactic agent and has in vitro antineoplastic activity, while adenosine functions as an immunosuppressor and is associated with neoangiogenesis, vasculogenesis, and evasion from the immune system. Inhibitors of ecto-5'-NT can prevent tumor progression, lower adenosine concentrations, and impede cancer growth by preventing immune system evasion.

UTP increases wound healing in the self assembled skin substitute (SASS)

Summary: In this study, the impact of purinergic receptor agonists on wound healing was investigated using self-assembled skin substitutes (SASS). The results showed that treatment with UTP accelerated wound healing, increased proliferation of keratinocytes and fibroblasts, and did not affect skin quality. Furthermore, the presence of ectoenzymes NTPDase1 and NPP1 in the wounded skin suggests their involvement in the wound healing process. Although these results are preliminary, they highlight the importance of understanding the role of extracellular nucleotides in skin healing and the need for further experiments to determine the underlying mechanisms.

Synthesis and biological evaluation of sulfamate derivatives as inhibitors of carbonic anhydrases II and IX

Summary: Inhibition of carbonic anhydrases is being explored as a potential treatment option for several pathological conditions. This study synthesized a series of 4-benzamidophenyl sulfamate derivatives and tested their inhibitory activity against two isoforms of carbonic anhydrase. The synthesized compounds showed potent inhibition of both isoforms at micromolar concentrations, with compound 1n and 1f being the most potent inhibitors for CA II and CA IX, respectively. Molecular modeling studies provided insights into the interactions between these compounds and the active sites of the enzymes.

Stomach-derived human insulin-secreting organoids restore glucose homeostasis

Summary: Huang et al. developed a method to generate pancreatic islet-like organoids from human gastric stem cells, which can restore glucose homeostasis in diabetic mice. This study provides a promising approach for cell therapy in diabetes by differentiating gastric stem cells into insulin-secreting cells that resemble beta-cells. The transplanted organoids successfully restored glucose balance for over 100 days in diabetic mice.

Characterization of purinergic signaling in tumor-infiltrating lymphocytes from lower- and high-grade gliomas

Summary: Malignant gliomas are aggressive and invasive tumors with a poor prognosis. The interactions between glioma cells and other components of the tumor microenvironment pose a challenge for effective treatment strategies. This study explores the purinergic signaling and ectoenzymes involved in the metabolism of extracellular nucleotides in the glioma microenvironment, with a focus on lower-grade gliomas and the differences between IDH-Mut and IDH-WT gliomas. The findings suggest that the increased ectoenzymes in the tumor microenvironment contribute to immunosuppression.

Silencing NTPDase3 activity rehabilitates the osteogenic commitment of post-menopausal stem cell bone progenitors

Inhibiting NTPDase3 or silencing its gene expression can restore the osteogenic potential of these cells.

Summary: The study discovered that heparin and its derivatives are potent and selective allosteric inhibitors of the poorly investigated ectonucleotidase NPP1. Synthesis and biological evaluation of sulfamoyl benzamide derivatives as selective inhibitors for h-NTPDases

Summary: The aim of this research is to synthesize sulfamoyl-benzamides as selective inhibitors of h-NTPDases. Various sulfonamides and carboxamides with biologically relevant substituents were synthesized and screened for their inhibitory activity against h-NTPDases isoforms. The most potent inhibitors were identified for each isoform, and molecular docking studies showed significant interactions with the target proteins.

The Application of Static Magnetic Stimulation Reduces Survival of SH-SY5Y Neuroblastoma Cells

Summary: The viability of SH-SY5Y neuroblastoma cells was reduced after 24 h and 6 days of exposure to SMS (p<0.05), while there were no differences for the other time points (p>0.05). However, this effect was not associated with cell death or cell cycle arrest (p>0.05). In contrast, the viability of HMV-II melanoma cells, used as a control, was not affected. Additionally, stimulated SH-SY5Y cells showed a decrease in mitochondrial mass at both exposure times and a reduction in autophagy and cell proliferation after 6 days (p<0.05).

Summary: This study investigated the expression of CD39 and CD73 enzymes in prostate cancer and their potential as therapeutic targets. The results showed higher levels of CD39 expression compared to CD73 in tissue samples and a correlation with Gleason score. In blood samples, CD39 expression in extracellular vesicles was significantly increased and positively correlated with ADP hydrolysis and Gleason score.
