Kaempferol-3-O-beta-D-glucuronide (K3G) having various pharmacological effects was explored for its anti-inflammatory effect in LPS induced RAW 264.7 cells and mice model. K3G significantly inhibited various pro-inflammatory mediators like IL-1 beta, NO, PGE2, and LTB4. It upregulated the secretion of anti-inflammatory cytokine IL-10. K3G is found to reduce inflammation when studied for parameters like phagocytic index, carrageenan induced paw edema in mice and organ weight. It reduced inflammation in a dose dependent manner both in-vitro and in-vivo. Further molecular insights into the study reveal that K3G blocks the phosphorylation of NF-kappa B which is key regulator of inflammation, thereby exhibiting anti-inflammatory potential. Hence, this study permits further investigation to develop K3G as anti-inflammatory drug.

Summary: UDP-N-acetylglucosamine enolpyruvyl transferase (MurA) is an important enzyme involved in bacterial cell wall synthesis. This study identified two potent inhibitors of MurA through virtual screening and validated their efficacy through in vitro assays. The study also discovered a novel binding site and revealed the mechanism of enzyme inhibition through conformational changes.

Study of potent CDK7 inhibitor secondary metabolites from Tecomella undulata

Setomimycin as a potential molecule for COVID-19 target: in silico approach and in vitro validation

Summary: This study investigates the potential of setomimycin as a therapeutic drug for COVID-19. Pure setomimycin was isolated from Streptomyces sp. RA-WS2 strain, and its in silico and in vitro anti-SARS-CoV-2 activity was evaluated. The compound was found to target the main protease (M-Pro) of SARS-CoV-2, inhibiting its dimerization. Additionally, setomimycin exhibited anti-inflammatory and anti-oxidant properties, suggesting its potential application against COVID-19 infections.

Mesenchymal stem cells attenuate systemic lupus erythematosus by inhibiting NLRP3 inflammasome activation through Pim-1 kinase

Summary: The activation of the inflammasome plays a critical role in the pathogenesis of systemic lupus erythematosus (SLE), and mesenchymal stem cells (MSC) have been shown to alleviate SLE by suppressing inflammasome activation. This study found that the NLRP3 inflammasome was activated in macrophages from SLE patients and mice, and its activation correlated with disease activity. After MSC transplantation, the severity of SLE was reduced, and NLRP3 inflammasome activation was inhibited. These findings suggest that MSC suppress inflammasome activation and provide a potential therapeutic target for SLE.
