Summary: Vibrio alginolyticus, an important pathogen in fish and humans, expresses a type III secretion system (T3SS) crucial for virulence. A chaperone protein gene vscB was cloned and a mutant strain HY9901ΔvscB was constructed, exhibiting decreased virulence to fish. Transcriptome sequencing identified 95 differentially expressed genes, with 57 up-regulated and 38 down-regulated in the ΔvscB strain. Quantitative RT-PCR analysis showed altered expression of T3SS-related genes in the ΔvscB strain. Vaccination with HY9901ΔvscB induced immune responses in pearl gentian grouper, suggesting it as a potential attenuated vaccine against V. alginolyticus infection.

Summary: The study evaluated the effectiveness of the Delta dctP strain as a live-attenuated vaccine against vibriosis caused by Vibrio alginolyticus in pearl gentian grouper. The expression of EcHsp40 in Epinephelus coioides is significantly up-regulated in response to bacterial or viral challenges, suggesting its involvement in pathogenic stimulation-induced apoptosis and viral replication.

Summary: In this study, the fliR deletion mutant Delta fliR of V.alginolyticus was constructed and evaluated, and the gene expression differences between wild-type and Delta fliR were analyzed by transcriptomics. Results showed that Delta fliR significantly affected cell motility and related pathways. Additionally, Delta fliR as a live attenuated vaccine demonstrated high protective efficacy and immune stimulation in groupers.

Summary: This study expressed and purified the Lrp recombinant protein from V. alginolyticus, which showed high antigenicity and can be used as an antigen for a subunit vaccine. It was found that chitosan oligosaccharide (COS) as an adjuvant could boost the immunological protection of the vaccine. The Lrp+COS subunit vaccine showed superior immunological impact and higher immune protection rates compared to the Lrp group.
