Summary: This study reveals the significant role of SPC25 in hepatocellular carcinoma (HCC) progression and its correlation with disease severity and poor survival. Mechanistically, SPC25 promotes DNA damage and activates the DNA-PK/Akt/Notch1 signaling cascade, leading to the regulation of HCC cell proliferation and stemness program. These findings provide potential biomarkers for predicting HCC progression and poor survival, as well as therapeutic targets for HCC patients.

Summary: Recent research has identified a novel lncRNA Linc-KILH, which is significantly up-regulated in HCC tissues and positively correlated with tumor size, microvascular invasion, intrahepatic metastasis and decreased survival in HCC patients. Silencing Linc-KILH can inhibit the proliferation and metastasis abilities of KRT19-positive HCC cells. Mechanistically, Linc-KILH interacts with KRT19 and inhibits the phosphorylation of KRT19, leading to enhanced translocation of KRT19 in HCC cells.

Summary: This study reveals the effect of TET3-mediated hydroxymethylation of DNA on the polarization of Kupffer cells (KCs) and its connection to the development of hepatocellular carcinoma (HCC). Knockdown of TET3 suppresses the viability, migration, and invasion of cocultured HCC cells and inhibits tumorigenesis in nude mice. The study also demonstrates that TET3 mediates the hydroxymethylation of ZMIZ1 promoter, resulting in M2 skewing of KCs through the Notch1/c-Myc axis.

Summary: This study characterized the molecular features of hepatocellular carcinoma (HCC) in Taiwan using a newly developed classification system based on the expression of Wnt-Hippo signaling pathway-related genes. The study identified four subclasses of HCC and analyzed their characteristics in terms of gene expression, clinical features, and prognosis. The study also identified signaling pathway-related genes that may influence immune cell infiltration. This novel molecular classification system may provide new insights for the treatment of HCC.
