Species Diversity and Functional Prediction of Surface Bacterial Communities on Aging Flue-Cured Tobaccos

Microbes on aging flue-cured tobaccos (ATFs) improve the aroma and other qualities desirable in products. Understanding the relevant organisms would picture microbial community diversity, metabolic potential, and their applications. However, limited efforts have been made on characterizing the microbial quality and functional profiling. Herein, we present our investigation of the bacterial diversity and predicted potential genetic capability of the bacteria from two AFTs using 16S rRNA gene sequences and phylogenetic investigation of communities by reconstruction of unobserved states (PICRUSt) software. The results show that dominant bacteria from AFT surfaces were classified into 48 genera, 36 families, and 7 phyla. In addition, Bacillus spp. was found prevalent on both ATFs. Furthermore, PICRUSt predictions of bacterial community functions revealed many attractive metabolic capacities in the AFT microbiota, including several involved in the biosynthesis of flavors and fragrances and the degradation of harmful compounds, such as nicotine and nitrite. These results provide insights into the importance of AFT bacteria in determining product qualities and indicate specific microbial species with predicted enzymatic capabilities for the production of high-efficiency flavors, the degradation of undesirable compounds, and the provision of nicotine and nitrite tolerance which suggest fruitful areas of investigation into the manipulation of AFT microbiota for AFT and other product improvements.

Summary: Three new cytochalasins, talachalasins A-C (1-3), were discovered from a deep-sea-derived fungus called Talaromyces muroii sp. SCSIO 40439. The structures and absolute configurations of 1-3 were determined using various analytical techniques. Talachalasins A-C (1-3) are unique cytochalasans with a 168-methyl group, which distinguishes them from over 500 known CYTs. Talachalasin C (3) is a novel 20,21-seco-cytochalasans with a 2-oxabicyclo-[3.3.1]nonan-3-one unit. Talachalasin A (1) exhibited moderate cytotoxic activity against four tumor cell lines (IC50: 3.40-10.02 μM). Talachalasin B (2) demonstrated comparable anti-RSV and HSV-1 activities (IC50: 12.5 p.M or IC50: 20.0 μM).

Enhancing thermal conductivity and chemical protection of bacterial cellulose/silver nanowires thin-film for high flexible electronic skin

Structural Characterization, Antioxidant and Antitumor Activities of the Two Novel Exopolysaccharides Produced by Debaryomyces hansenii DH-1

Summary: Exopolysaccharides produced by edible microorganisms, such as S1 and S2, have excellent physicochemical properties and significant biological activity, making them advantageous for the food and pharmaceutical industries. Both S1 and S2 exhibited good thermal stability and potent hydroxyl radical scavenging activity, with S1 also possessing strong iron-reducing capacity. In vitro antitumor assays showed that S1 and S2 significantly inhibited the proliferation of Hela, HepG2, and PC-9 cancer cells, with PC-9 being more sensitive to S1. These findings suggest that S1 and S2 have great potential as natural antioxidants and candidates for cancer treatment in the food and pharmaceutical industries.

Totopotensamide Congeners from a Halogenase-Inactivated Mutant

Summary: The installation of halogen atoms into aromatic and less activated polyketide substrates by halogenases is a powerful strategy to tune the bioactivity, bioavailability, and reactivity of compounds. In this study, the halogenase TotH was confirmed to catalyze the C-4 chlorination in the biosynthetic pathway of totopotensamide A (1), leading to the formation of ClMeDPG. Six new deschloro totopotensamide (TPM) congeners TPMs H2-H7 (5-10) were isolated and their structures were elucidated. Additionally, the cytotoxicity of compounds 5, 6, 8, and 9 against several cell lines was evaluated.

Inconsistent stoichiometry and growth responses of two coexisting dominant species to various N and P supplies in a supratidal wetland of the Yellow River Delta

Summary: The availability and stoichiometry ratio of nitrogen (N) and phosphorus (P) are important factors in plant trophic dynamics and primary production. However, the response of supratidal wetland herbs to varying N and P supplies is not well understood. In this 4-year field manipulation experiment, the aboveground biomass, leaf N and P concentrations, and N:P ratios of two dominant herbs were examined under different nutrient supply levels and N:P supply ratios. The results showed that the leaf trophic dynamics and aboveground biomass of the herbs were influenced by both overall supply level and N:P supply ratio.

Biosynthetic Diversification of Fidaxomicin Aglycones by Heterologous Expression and Promoter Refactoring

Summary: In this study, an alternative biological route to produce structurally diverse fidaxomicin aglycones was developed by refactoring the promoter and expressing it in a different host. The resulting titers of the five fidaxomicin aglycones ranged from 4.9 to 15.0 mg L-1. Overall, these findings demonstrate the potential of using a biological strategy to construct and diversify fidaxomicin aglycones.

Biosynthesis and Engineered Overproduction of Everninomicins with Promising Activity against Multidrug-Resistant Bacteria

Summary: Ribosome-targeting oligosaccharides, everninomicins (EVNs), are promising drug leads with a unique mode of action. By duplicating the biosynthetic gene cluster (BGC) in Micromonospora sp. SCSIO 07395, the production of EVNs was enhanced, allowing for bioactivity evaluation. EVNs showed significant inhibition against multidrug-resistant bacteria, with potency comparable or superior to vancomycin, linezolid, and daptomycin. The BGC duplication strategy successfully increased the titer of bioactive EVN M from trace amounts to 98.6 mg L-1, demonstrating the utility of a bioengineering approach for enhanced production and chemical diversification of EVNs.

A pH-stable alkaline pectate lyase produced by the newly identified strain Bacillus altitudinis CAS-WZS-08

Summary: This study aimed to obtain a strain capable of producing pectate lyase with good pH stability. Bacillus altitudinis CAS-WZS-08, producing pectate lyase with good pH stability, was isolated. The optimal fermentation conditions for pectate lyase production were determined, and the purified protein was identified as pectate lyase with excellent juice extraction ability.

Anti-oxidative and anti-aging effects of mannoprotein-rich yeast cell wall enzymatic hydrolysate by modulating gut microbiota and metabolites in Caenorhabditis elegans

Summary: In this study, the antioxidant and anti-aging activities of mannoprotein-rich yeast cell wall enzymatic hydrolysate (MYH) were investigated using the Caenorhabditis elegans (C. elegans) model. The results showed that MYH could increase the lifespan and anti-stress ability of C. elegans by enhancing the activity of antioxidant enzymes and decreasing levels of oxidative stress and apoptosis. Additionally, MYH was found to modulate the expression of specific mRNAs related to antioxidant and anti-aging pathways. Furthermore, MYH was shown to improve the gut microbiota composition and metabolite levels in C. elegans. This study contributes to understanding the antioxidant and anti-aging activities of microorganisms and the development of functional foods.

Design of Ka-Band Phased Array Antenna with Calibration Function

Summary: In this paper, a novel structure of Ka-band based phased array antenna with calibration function is proposed. The antenna adopts the active phased array system and works in the dual polarization separation mode. The schematic diagram and design details of the Ka-band antenna are provided. The transceiver network and calibration network are discussed, and simulation and measurement results are shown.
