Summary: This review discusses synthetic methods for obtaining 1,2,4-oxadiazole and their application in pharmaceutical preparation. The methods are divided into three groups, including two-stage protocols, one-pot synthesis, and oxidative cyclization. These methods have significant potential in drug design, allowing for the preparation of thermosensitive 1,2,4-oxadiazole and expanding the use of oxadiazole core as a linker in bioactive compound design.

Summary: The aerobic visible-light induced intermolecular S-N bond construction without the addition of photosensitizer, metal, or base allows for the synthesis of 1,2,4-thiadiazoles from thioamides. The preliminary mechanistic investigation indicates that the excited state of thioamides undergoes a single-electron-transfer process to form thioamidyl radicals, which can be further converted into 1,2,4-thiadiazoles through desulfurization and oxidative cyclization. This reaction with good functional group tolerance represents a green method for the construction of S-N bonds.

Summary: A new COF based on imine bonds with dual-pore structure and high crystallinity was successfully synthesized, and selectively metallated with Pt on the N donor in the larger pores. The metallated COF showed excellent performance as a recyclable heterogeneous photocatalyst for decarboxylative difluoroalkylation and oxidative cyclization reactions.

Summary: Here, we report a generally applicable method for preparing N-substituted benzo[4,5]thiazolo[2,3-c][1,2,4]triazol-1-ium salts from air-stable precursors. This method involves the selective deprotection of a para-methoxybenzyl protected thiol, followed by C-H functionalization of the linked 1,2,4-triazolium salts under oxidative conditions. The synthesized tricyclic thiazolium salts can contain various electron-withdrawing and electron-donating aromatic substituents, as well as aliphatic substituents, and are tolerant to many functional groups.

Summary: A metal-free oxidative cyclization process using trifluoroacetimidohydrazides and D-glucose has been developed for the efficient assembly of 3-trifluoromethyl-1,2,4-triazoles. D-glucose serves as the C1 synthon to provide the methine source in the reaction. Control experiments were conducted to elucidate the reaction mechanism, and the synthetic utility of the method was demonstrated through scale-up reactions and the synthesis of a key skeleton for an NKI-receptor ligand.

Summary: An efficient method for the synthesis of alpha-(1,3,5-triazinylthio)-ketones from 1,3-dicarbonyl compounds with 1,3,5-triazine-2-thiols has been developed. The reaction involves C-C bond cleavage and C-S bond reconstruction of 1,3-dicarbonyl compounds, and can tolerate beta-keto esters, beta-keto amides, and 1,3-diones. In addition, the annulation of 1,3,5-triazine-2-thiols with chalcones has been achieved for the synthesis of thiazolo-[3,2-a]-[1,3,5]-triazines in moderate to good yields, and with a broad functional group tolerance.

Summary: A convenient and efficient method for the synthesis of 3,5-disubstituted 1,2,4-oxadiazoles from N-benzyl amidoximes was developed using anodic oxidation to generate the iminoxy radical. The transformation involves 1.5-Hydrogen Atom Transfer (1,5-HAT) and intramolecular cyclization, providing a facile and practical way for the preparation of 1,2,4-oxadiazoles with simple operation, mild conditions, broad substrate scope, and high functional group compatibility.
