Valproic acid induces three novel cytotoxic secondary metabolites in Diaporthe sp., an endophytic fungus from Datura inoxia Mill.

Addition of the valproic acid (histone deacetylases inhibitor) to a culture of an endophytic fungus Diaporthe sp. harbored from Datura inoxia significantly altered its secondary metabolic profile and resulted in the isolation of three novel compounds, identified as xylarolide A (1), diportharine A (2) and xylarolide B (3) along with one known compound xylarolide (4). The structures of all the compounds (1-4) were determined by detailed analysis of ID and 2D NMR spectroscopic data. The relative configurations of compounds 1-3 were determined with the help of NOESY data and comparison of optical rotations with similar compounds with established stereochemistry. All the isolated compounds were screened for antibacterial, antioxidant and cytotoxic activities. Xylarolide A (1) and xylarolide (4) displayed significant growth inhibition of MIAPaCa-2 with an IC50 of 20 and 32 mu M respectively and against PC-3 with an IC50 of 14 and 18 mu M respectively. Moreover, compound 1 displayed significant DPPH scavenging activity with EC50 of 10.3 mu M using ascorbic acid as a positive control. (C) 2018 Elsevier Ltd. All rights reserved.

Serine-glycine-betaine, a novel dipeptide from an endophyte Macrophomina phaseolina: isolation, bioactivity and biosynthesis

Summary: This study isolated a novel dipeptide, serine-glycine-betaine, from the endophyte Macrophomina phaseolina associated with Brugmansia aurea L., which exhibited activity against human cancer cells by inducing apoptosis. Genomic analysis revealed the potential biosynthesis pathway of serine-glycine-betaine. The findings suggest dipeptides like serine-glycine-betaine and tyrosine-betaine may be specific to fungal genera and could be used for diagnostic purposes.

Mutation, Chemoprofiling, Dereplication, and Isolation of Natural Products from Penicillium oxalicum

Summary: Diethyl sulfate (DES)-based chemical mutagenesis was used to diversify secondary metabolites in different fungal strains. A mutant strain of Penicillium oxalicum was isolated and found to produce 10 bioactive molecules. Compounds like tuckolide, methylpenicinoline, and penipanoid A showed potent antibacterial activity, while N-[2-(4-hydroxyphenyl)ethenyl]formamide and tuckolide demonstrated strong antioxidant properties. DES-based mutagenesis was proven to be an effective strategy in diversifying fungal secondary metabolites.

Biological Activity of Picrorhiza kurroa: A Source of Potential Antimicrobial Compounds against Yersinia enterocolitica

Summary: Yersiniosis, a zoonotic disease caused by Yersinia enterocolitica, is prevalent in Europe and known for its high antibiotic resistance. This study evaluated the antimicrobial activity of an extract from Picrorhiza kurroa against Yersinia enterocolitica. The extract showed effective antimicrobial activity and potential bioactive compounds were identified. Picroside-1 exhibited higher antimicrobial potential and lysed Yersinia enterocolitica cells. Docking analysis revealed its interaction with the dihydrofolate reductase protein, suggesting it as a possible drug candidate for Yersinia enterocolitica.

Design of MERS-CoV entry inhibitory short peptides based on helix-stabilizing strategies

Summary: This study successfully developed a short peptide mimetic as a MERS-CoV fusion inhibitor by reproducing the key recognition features of the HR2 helix. The resulting 23-mer lipopeptide showed comparable inhibitory effect to the 36-mer HR2 peptide HR2P-M2. This has important implications for developing short peptide-based antiviral agents to treat MERS-CoV infection.

Conformationally constrained potent inhibitors for enhancer of zeste homolog 2 (EZH2)
