Circulating tumor cells (CTCs) shed by the primary tumor and metastases are considered a real-time liquid biopsy', reflecting the disease complexity that evolves during progression, showing in its late stages different genetic, epigenetic and expression features. Consequently, heterogeneity and development of characteristic features upon disease progression are the two main goals that emerging technologies should account for in view of a clinical application. Single-cell analysis, now possible due to technological advances, may help elucidate tumor heterogeneity at the CTC level. This review focuses on the necessary steps for the analysis of CTCs at the single-cell level. A concise overview is given on the alternative methods referring in particular to studies on the mutational status of single CTCs from cancer patients.

Supervised learning methods for the recognition of melanoma cell lines through the analysis of their Raman spectra

Summary: Malignant melanoma is a deadly form of skin cancer caused by genetic mutations in melanocytes, with BRAF and NRAS being the most frequent genes involved. This study utilized Raman spectroscopy to examine melanoma cell lines with specific mutations, developing accurate classification algorithms based on the obtained molecular information. The results suggest that Raman spectroscopy could be an effective tool for detecting molecular differences between melanoma mutations, with high accuracy in discriminating different cell types.

Summary: Liquid biopsy, including cfDNA and CTCs, shows prognostic value in KRAS mutated mCRC. CTCs are a negative prognostic marker at baseline, while cfDNA is easily accessible for monitoring tumor mutations over time. In longitudinal monitoring, these two markers complement each other in assessing disease progression.

Summary: This study investigated the predictive value of miRNA and ARV7 mutational status in mCRPC patients, as well as the role of CTC detection in 1st line therapy. Results showed that CTC+ and ARFL+ patients had significantly reduced TTCR, indicating the important impact of CTC detection and ARFL expression on patient prognosis.
