The results suggest that baseline serum levels of BAFF are the best predictor of response to Belimumab, confirming previous study findings and indicating the utility of this biomarker for identifying responsive patients.

Impact of Δ9-Tetrahydrocannabinol on Rheumatoid Arthritis Synovial Fibroblasts Alone and in Co-Culture with Peripheral Blood Mononuclear Cells

Summary: This study reveals that delta 9-Tetrahydrocannabinol (THC) has both pro-inflammatory and anti-inflammatory effects on synovial fibroblasts from patients with rheumatoid arthritis (RASF) and peripheral blood mononuclear cells (PBMC) from healthy donors. The effects of THC depend on the activating stimulus and concentration. Consequently, THC might be a potential treatment for inflammation in rheumatoid arthritis, but the optimal concentration needs to be determined.

Anti-Inflammatory Effects of Cannabigerol in Rheumatoid Arthritis Synovial Fibroblasts and Peripheral Blood Mononuclear Cell Cultures Are Partly Mediated by TRPA1

Summary: Since its legalization, cannabis preparations containing CBD and THC have been used by RA patients for pain and inflammation relief. However, the minor cannabinoid CBG, which also has anti-inflammatory properties, is not widely used due to lack of studies. CBG binds to cellular target proteins, including TRPA1, and can modulate disease activity in RA by reducing oxidative stress and pro-inflammatory pathways. CBG showed anti-inflammatory effects by increasing calcium levels and enhancing drug uptake in stimulated RASF cells, decreasing cell viability and IL-6/IL-8 production. In PBMC cultures, CBG modulated cytokine and immunoglobulin production, partially mediated by TRPA1. CBG could be used as add-on therapy in RA to reduce IL-6 and autoantibody levels.

Components of the sympathetic nervous system as targets to modulate inflammation - rheumatoid arthritis synovial fibroblasts as neuron-like cells?

Summary: This study found that rheumatoid arthritis synovial fibroblasts possess a complete and functional catecholamine machinery, which is altered under inflammatory conditions. These results shed further light on the involvement of sympathetic neurotransmitters in rheumatoid arthritis pathology and might open therapeutic avenues to counteract inflammation with the MAO enzymes being key candidates.

Understanding these risk mechanisms may offer prevention and treatment opportunities for insomnia in RA, potentially improving pain and disease outcomes.
