Doublecortin (DCX) is a microtubule-associated protein that has been considered a marker for neuronal precursors and young migrating neurons during the development of the central nervous system and in adult neurogenic niches. The retina of fishes represents an accessible, continuously growing and highly structured (layered) part of the central nervous system and, therefore, offers an exceptional model to extend our knowledge on the possible role of DCX in promoting neurogenesis and migration to appropriate layers. We have analyzed the distribution of DCX in the embryonic and postembryonic retina of a small shark, the lesser spotted dogfish Scyliorhinus canicula, by means of immunohistochemistry. We investigated the relationship between DCX expression and the neurogenic state of DCX-labeled cells by exploring its co-localization with the proliferation marker PCNA (proliferating cell nuclear antigen) and the marker of neuronal differentiation HuC/D. Since radially migrating neurons use radial glial fibers as substrate, we explored the possible correlation between DCX expression and cell migration along radial glia by comparing its expression with that of the glial marker GFAP (glial fibrillary acidic protein). Additionally, we characterized DCX-expressing cells by double immunocytochemistry using antibodies against Calbindin (a marker for mature bipolar and horizontal cells in this species) and Pax6, which has been proposed as a regulator of cell proliferation, cell differentiation, and neuron diversification in the neural retina of sharks. Strong DCX immunoreactivity was observed in immature cells and cell processes, at a time when retinal cells were not yet organized into different laminae. DCX was also found in subsets of mature ganglion, amacrine, bipolar and horizontal cells long after they had exited the cell cycle, a pattern that was maintained in juveniles and adults. Our results on DCX expression in the retina are compatible with a role for DCX in cell migration within the immature retina, and in dynamic neuronal plasticity in the mature retina. We also provide evidence of DCX expression in discrete cells in the retinal pigment epithelium of prehatching embryos and juveniles, which suggest that retinal pigmented epithelial cells in sharks, as in mammals, have an intrinsic capacity to proliferate and differentiate into cells with neural identity. Summary: Neurogenesis is the process of generating new neurons by progenitor cells, which is restricted to specific neurogenic niches. The retina of cartilaginous fish such as catsharks shows active proliferation and neurogenesis in juveniles but a reduction in cell proliferation after sexual maturation. RNA-Seq analysis revealed a decrease in gene expression related to cell proliferation and neurogenesis in adult catsharks, providing valuable insight into signaling pathways controlling neurogenesis in vertebrate retinas.

Glial markers were highly expressed in the olfactory nerve layer and the glomerular layer during the analyzed developmental stages. These findings provide insights for future studies on the differential roles of glial cells in the embryonic development and adulthood of catshark olfactory bulb.

Decline in Constitutive Proliferative Activity in the Zebrafish Retina with Ageing

Embryonic nutritional hyperglycemia decreases cell proliferation in the zebrafish retina

Summary: Diabetic retinopathy is a major cause of blindness worldwide. Studies have shown that hyperglycemia during early retinal development can lead to decreased cell numbers and altered cell differentiation. This study on zebrafish embryos found that hyperglycemia increases cell death and decreases cell proliferation in the developing retina, which could contribute to the decline in mature retinal cells.

Summary: This study successfully addressed the limitations of resveratrol, an antioxidant agent, by preparing single and mixed micelles of Pluronic (R) F127 and casein. These micelles significantly increased the solubility of resveratrol, remained stable, and exhibited potent antioxidant properties. They also showed antimicrobial activity. However, mixed micelles displayed lower permeability through the sclera compared to single micelles.

Summary: This study aimed to examine the intraocular tolerability of cetuximab when applied intravitreally, and its effect on axial elongation. The results showed that intravitreal injections of cetuximab could reduce axial elongation in a dose-dependent and number of treatment-dependent manner, without causing intraocular toxic effects.

Summary: Aniridia is a panocular condition characterized by the loss of iris, caused mainly by mutations in the PAX6 gene. This study assessed the effects of Pax6-haploinsufficiency on retinal morphology and vision in Pax6Sey mice. Summary: The sodium iodate (NaIO3) model is a valuable tool in studying dry AMD and evaluating the retinal structure. Through extensive research, it has been found that the death of RPE cells caused by NaIO3 injection is closely related to vision loss in patients with dry AMD. Therefore, studying NaIO3 can provide insights into the mechanisms of RPE and photoreceptor cell death and contribute to understanding RPE degeneration in AMD.

A new model of axon degeneration in the mouse optic nerve using repeat intraocular pressure challenge

Summary: We present a new experimental model for inducing retinal ganglion cell (RGC) dysfunction and degeneration in mice. This model allows for the reliable induction of RGC dysfunction and loss, with the resilience of the optic nerve being dependent on the age of the animal. The time-defined nature of the functional and structural changes observed in this model will facilitate the study of glial responses and the evaluation of neuroprotective treatments after intraocular pressure (IOP) injury.
