Summary: This study developed a mathematical kinetic model to explain the change in volume of injected dermal fillers, separating the kinetics of the fillers into hydration, injection-site reaction, and enzyme-mediated degradation. The model was based on a two-compartment system, with one compartment for injection and another for observation of swelling and degradation. The rate-limiting step for complete degradation of the fillers in vivo was found to be the swelling phase, as indicated by the swelling rate constant (K-swell), providing insight for scientists developing new dermal fillers.

Summary: Hyaluronic acid is an important ingredient in dermal fillers that is often cross-linked to improve properties and durability. Poly(ethylene glycol) diglycidyl ether (PEGDE) has been introduced as a crosslinker with similar chemical reactivity as the commonly used crosslinker BDDE. However, there are no literature methods available to monitor PEGDE content. This study presents an HPLC-QTOF method for routine examination of PEGDE content in HA hydrogels, following the guidelines of the International Council on Harmonization.
