Summary: This study evaluated the role of toll-like receptor 3 (TLR3) activating RNA in the pathogenesis of osteoarthritis (OA). The results showed that RNA released by apoptotic chondrocytes can exacerbate inflammatory cartilage changes by activating TLR3 signaling. Inhibition of TLR3 could be a potential therapeutic target for OA treatment.

In this study, a novel circRNA, Circ_0088194, was found to be upregulated in RA fibroblast-like synoviocytes (RA-FLSs) and promote invasion and migration by acting as a sponge for miR-766-3p to regulate the expression of MMP2. This study suggests that Circ_0088194 may be a potential therapeutic target for preventing and treating RA.

Summary: This study reveals that G protein-coupled receptor neurokinin-1 (NK1R) is significantly upregulated in lung cancer samples and is associated with advanced clinical stages and poor prognosis. NK1R co-expresses with epidermal growth factor receptor (EGFR) and interacts with it in tumor cells. Activation of NK1R promotes lung cancer cell proliferation and migration through EGFR signaling pathways. Inhibition of NK1R suppresses cell proliferation and migration, and knockdown of NK1R slows down tumor growth. The presence of a selective NK1R antagonist enhances the sensitivity of lung cancer cells to specific drugs.

This study demonstrates that there is increased expression of HB-EGF in IPF patients and mice with pulmonary fibrosis, and HB-EGF plays a critical role in regulating fibrosis progression.

Summary: Recent studies suggest that introducing metal-binding non-canonical amino acids (NCAAs) into the N-terminal domain of TIMP2 can improve its specificity towards MMP-2 and MMP-9, resulting in increased inhibitory potency. These NCAA variants showed a significant loss of inhibitory activity towards MMP-14 but not towards MMP-2 and MMP-9. Molecular modeling revealed that the bulky NCAA substituents can be better accommodated at the intermolecular interface with MMP-2 and MMP-9, forming stabilizing polar interactions.
