IL-36α Regulates Tubulointerstitial Inflammation in the Mouse Kidney

IL-36 alpha, a member of the IL-1 family, is a crucial mediator of inflammatory responses. We previously found that IL-36 alpha was overexpressed in injured distal tubules (DTs); however, its pathological function remains unclear. Herein, unilateral ureter obstruction (UUO) or folic acid (FA) injection was performed in mouse kidneys to assess the role of IL-36 alpha in kidney injury. IL-36 alpha mRNA and protein expression significantly increased in the kidneys within 24 h after UUO. IL-36 alpha localized to dilated DTs. IL-36 alpha expression significantly correlated with the progression of tubulointerstitial cell infiltration and tubular epithelium cell death in UUO kidneys and with renal dysfunction in FA-induced acute kidney injury mice. At 24 h after UUO, IL-36 alpha(+) DT epithelial cells showed loose intercellular digitations. IL-1RL2, an IL-36 alpha receptor protein, localized to podocytes, proximal tubules, and DTs in the healthy kidney. IL-1RL2 was expressed in interstitial cells and platelets or extended primary cilia of DT epithelial cells in UUO kidneys. IL-36 alpha stimulation promoted the production of IL-6 and Prss35, an inflammatory cytokine and collagen remodeling-associated enzyme, respectively, in cultured NIH3T3 fibroblasts. UUO-treated IL-36 alpha-knockout (KO) mice showed milder kidney injury features than wild-type (WT) mice did. In UUO kidneys from IL-36 alpha-KO mice, the expression of genes associated with inflammatory response and sensory perception was significantly different from that in WT mice. Altogether, our data indicate an association between intrarenal IL-36 alpha overexpression and the progression of tubulointerstitial inflammations and morpho-functional alterations of DT epithelial cells. IL-36 alpha may be a novel kidney injury marker useful for evaluating DT damages.

Expression of Indian hedgehog signaling in murine oviductal infundibulum and its relationship with epithelial homeostasis

Summary: In this study, we investigated the expression of hedgehog signaling-related components in the murine oviductal infundibulum and found that the homeostasis of the infundibulum epithelium was disturbed in MRL/lpr mice. The motile cilia of ciliated epithelial cells in the infundibulum harbored Hh signaling pathway-related molecules. However, these molecules were significantly downregulated in MRL/lpr mice at 6 months of age. Pax8 and Foxj1 expression levels were positively correlated with those of Ptch1, Smo, and Gli2. Overall, Hh signaling is involved in the regulation of ciliated epithelium homeostasis in the infundibulum and interacts with inflammation, leading to disturbances in epithelial homeostasis and reproductive function.

Parkinson's Disease Risk and Hyperhomocysteinemia: The Possible Link

Summary: Parkinson's disease (PD) is a degenerative brain disorder characterized by the loss of dopamine neurons in the substantia nigra and the accumulation of Lewy bodies and α-synuclein in the brain. Patients with PD often have deficiencies in vitamins such as folate, vitamin B6, and vitamin B12, leading to increased levels of homocysteine in the bloodstream, which may contribute to the development and progression of PD through oxidative and inflammatory pathways. Hyperhomocysteinemia is implicated in neurodegenerative disorders and triggers PD through various mechanisms, including oxidative stress, mitochondrial dysfunction, apoptosis, and endothelial dysfunction. Inflammatory signaling pathways like NF-kappa B and NLRP3 inflammasome are also involved in the pathogenesis of PD. In conclusion, hyperhomocysteinemia plays a role in the neuropathology of PD by directly inducing degeneration of dopamine neurons or indirectly activating inflammatory signaling pathways.

Oocyte cumulus complex quality and oviduct transportation velocity in systemic autoimmune disease model mice

Summary: Oocyte transportation in the oviduct is influenced by the interaction between ciliated epithelial cells and cumulus cells. In this study, we investigated the effect of cumulus-oocyte complex (COC) quality on the transportation property of COCs by comparing the transportation velocity of COCs (TVC) in different combinations of infundibula and COCs collected from various mice. Our results showed that the TVC was lower in aged MRL strains compared to aged C57BL/6N mice, indicating that aging affects COCs transportation in MRL strains. Moreover, the TVC was the lowest in aged MRL/MpJ-Fas (lpr/lpr) mice, while it accelerated when the infundibulum or COCs were collected from other strains, suggesting that the properties of COCs also play a role in determining their transportation property.

Microenvironmental Changes in Mediastinal Fat-associated Lymphoid Clusters and Lungs in Early and Late Stages of Metastatic Lung Cancer Induction

Summary: By comparing the changes in the lungs and mediastinal fat-associated lymphoid clusters (MFALCs) between patients with metastatic lung melanoma (MLM) and healthy individuals, it was found that MLM patients had a significant increase in immune cell populations in MFALCs, which may be associated with the progression of MLM. Therefore, targeting immune cells in MFALCs could be a promising therapeutic strategy for cancer treatment.

Transcriptome and proteome analysis of dogs with precursor targeted immune-mediated anemia treated with splenectomy

Summary: In this study, splenectomy was performed as an alternative therapy for refractory PIMA in dogs, and gene expression levels in the spleens and serum of dogs with PIMA were analyzed. Transcriptome analysis revealed differential gene expression in the spleens of dogs with PIMA, with up-regulation of genes such as S100A12, S100A8, and S100A9. Immunohistochemistry confirmed higher levels of S100A8/A9 protein expression in dogs with PIMA. Proteome analysis identified differentially expressed proteins in the serum samples before and after splenectomy, with up-regulation of proteins in the pre-splenectomy samples. Pathway analysis revealed the involvement of the lectin pathway of complement activation in the pre-splenectomy samples. These findings provide deeper insights into the pathology and mechanisms of splenectomy for PIMA.

Metataxonomic Analysis of the Uterine Microbiota Associated with Low Fertility in Dairy Cows Using Endometrial Tissues Prior to First Artificial Insemination

Summary: We examined the uterine microbiota associated with low fertility in dairy cows and found two new insights into their relevance to fertility. The deterioration in reproductive performance due to low fertility leads to significant economic losses on dairy farms, making the uterine microbiota an interesting area for research. Our study analyzed the uterine microbiota in dairy cows and found that it can vary depending on farm management practices, indicating its potential as a measure for low fertility.

Effects of autoimmune abnormalities on skeletal muscle regeneration after needle puncture in mice

Summary: Immune cells play a crucial role in skeletal muscle repair, and systemic autoimmune diseases can impair muscle regeneration. This study found that, regardless of the effects of autoimmune diseases, injured skeletal muscles caused by minor trauma can normally regenerate, although lymphocyte infiltrations were more severe in mice with autoimmune diseases.

Calbindin Has a Potential Spatiotemporal Correlation with Proliferation and Apoptosis in the Postnatal Rat Kidney

Summary: The study aimed to investigate the influence of proliferation and apoptosis in different compartments of the kidney on the developmental function of the protein calbindin-D28k. The results showed that calbindin localization was inversely related to PCNA and ssDNA in the nephron compartments, indicating their role in bone-building and muscle contraction efficiency during animal development.

Analysis of gene expression in poultry red mite, Dermanyssus gallinae, by RNAscope in situ hybridization

Summary: In this study, RNAscope-based in situ hybridization (ISH) was used to analyze gene expression in the poultry red mite (PRM). The results showed that Dg-CatD-1 mRNA was mainly expressed in the digestive tract, while Dg-Cystatin mRNA was expressed in the ovaries and digestive tract. RNAscope ISH is an effective method for investigating gene expression and characterizing target genes in PRM.

Histological Characteristics of Conjunctiva-Associated Lymphoid Tissue in Young and Adult Holstein Cattle
