lodixanol as a Contrast Agent in a Fibrin Hydrogel for Endodontic Applications

The application of biomaterials used in regenerative endodontics should be traceable. In this study, we checked some basic effects of rendering a fibrin hydrogel radiopaque using an iodine-based contrast agent (iodixanol) approved for systemic application. Fibrin hydrogels were prepared from a fibrin sealant (Tisseel) using either an isotonic iodixanol solution (Visipaque 320, test) or Tris buffer (control) as a diluent. Gelation kinetics, radiopacity, and swelling of lyophilized hydrogels were tested using standard methods. Hydrogel structure was evaluated using scanning electron microscopy (SEM). Furthermore, iodixanol release from the test gels was assessed using spectrophotometry, and tissue compatibility was compared between test and control hydrogels using the chick chorioallantoic membrane (CAM) assay. Results were compared using pairwise t -test, p < 0.05. lodixanol caused a 70-fold delay in gelation to 26 min in the test compared to the control hydrogels (22 +/- 1 s). Radiopacity of the test gels was 1.9 +/- 0.2 mm Al/mm, compared to zero in the control hydrogels. Lyophilized hydrogel swelling was strongly reduced when iodixanol was added to the hydrogel (p < 0.05). Test hydrogels had an altered SEM appearance compared to controls, and exhibited a reduced porosity. lodixanol release from the test hydrogels reached 14.5 +/- 0.5% after 120 h and then ceased. This release did not have any apparent toxic effect and neither affected the viability, nor the physiology or vascularization of the CAM of fertilized chicken eggs. lodixanol can render a fibrin hydrogel radiopaque and maintains its tissue compatibility, yet impacts gelation kinetics and hydrogel porosity.

Assessment of Irrigant Agitation Devices in Simulated Closed and Open Root Canal Systems

Summary: This study compared the in vitro efficacy and safety of different irrigant activation devices in apically closed and open root canal systems. The results showed that ultrasonic agitation was more effective in cleaning the central area of the root canal, while it had a lower effect in cleaning the simulated periapical lesion. Therefore, ultrasonic agitation was found to be more targeted and safer after the administration of sodium hypochlorite.

Nanocrystalline Apatites: Post-Immersion Acidification and How to Avoid It-Application to Antibacterial Bone Substitutes

Summary: Biomimetic nanocrystalline apatites analogous to bone mineral can be prepared using soft chemistry. However, immersion in aqueous media of dried non-carbonated biomimetic apatite crystals may generate an acidification event, which could limit their further development. In this study, the acidification phenomenon was explored, and a practical strategy to circumvent it was proposed. The non-cytotoxicity and antibacterial features of the post-equilibrated nanocrystalline apatites were demonstrated, suggesting their relevance in the field of antibacterial bone substitutes.

The Notch-mediated circuitry in the evolution and generation of new cell lineages: the tooth model

Summary: The Notch pathway is involved in cell fate specification and embryonic development. The Jagged2 gene, which encodes a ligand for Notch receptors, is expressed early in tooth development. Mutations in Jagged2 result in abnormal tooth morphology and impaired enamel deposition. The Notch signaling cascade is disrupted in Jagged2 mutant teeth, leading to dental structures more reminiscent of fish enamel. We propose that the increased number of Notch homologues in metazoa enabled the formation and maintenance of distinctive cell fates within organs and tissues during evolution.

Bioactive and Elastic Emulsion Electrospun DegraPol Tubes Delivering IGF-1 for Tendon Rupture Repair

Summary: Tendon injuries can result in limited range of motion and poor biomechanical outcomes. A novel three-layer tube based on polymer DegraPol with incorporated IGF-1 was developed using emulsion electrospinning. The IGF-1-containing tubes exhibited sustained release of the growth factor and showed bioactivity by upregulating gene expression. They also demonstrated superior mechanical properties compared to pure DP tubes. These tubes may help accelerate the healing process and reduce adhesion formation.

Influence of Scaffold Microarchitecture on Angiogenesis and Regulation of Cell Differentiation during the Early Phase of Bone Healing: A Transcriptomics and Histological Analysis

Summary: This study utilized additive manufacturing to produce two different diameter tricalcium phosphate-based bone scaffolds and observed them in vivo for 10 days followed by gene sequencing and histological analysis. The results showed upregulation of genes related to immune response, cell adhesion, and cell migration in both scaffolds. However, only the scaffold with a 0.50mm diameter showed significant overexpression of genes involved in angiogenesis, cell differentiation regulation, ossification, and bone development. Immunohistochemistry and high-resolution CT scans also revealed more blood vessels and mineralized tissue in the 0.50mm scaffold. Therefore, different filament diameters significantly influence angiogenesis and cell differentiation regulation in the early phase of bone regeneration, impacting the overall clinical outcome.

Notch Signaling Pathway in Tooth Shape Variations throughout Evolution

Summary: Evolutionary changes in vertebrates are often accompanied by genetic alterations that affect tooth crown shape. The Notch pathway, a highly conserved signaling pathway, plays a crucial role in morphogenetic processes, including tooth development. Experimental studies on developing mouse molars showed that the loss of the Notch-ligand Jagged1 leads to minor modifications in tooth crown shape, similar to those observed during the evolution of Muridae species. RNA sequencing analysis revealed that these alterations are caused by changes in the expression of over 2000 genes, and that Notch signaling interacts with other important morphogenetic networks, such as Wnts and Fibroblast Growth Factors. The findings from this study provide insights into the role of Notch/Jagged1-mediated signaling in dental variations during evolution.

Precision-milled simulated curved root canals in bovine dentine for the assessment of chemo-mechanical root canal preparation

Extracellular matrix remodelling in dental pulp tissue of carious human teeth through the prism of single-cell RNA sequencing
