The whitebacked planthopper Sogatella furcifera (Horvath) is an important pest of rice throughout Asia. Application of chemical insecticide is the main approach to suppress the field populations of S. furcifera. In this study, neonicotinoid insecticide resistance in field populations of S. furcifera were evaluated. The results showed that some field populations of S. furcifera had developed moderate level of resistance to imidacloprid (RR = 1.1-16.4), thiamethoxam (RR = 0.8-14.9), dinotefuran (RR = 1.2-16.6) and acetamiprid (RR = 3.3-12.2), low level of resistance to nitenpyram (RR = 1.1-9.5) and clothianidin (RR = 1.3-8.7) in Central China. Moreover, there were an increasing trend in neonicotinoid insecticide resistance in the period 2011-2015. The results of current study may form the basis to identify and evaluate the resistance tendency of S. furcifera against neonicotinoid insecticides, which could make effective management recommendations to avoid further development of insecticide resistance in S. furcifera.

miRNAs targeting CYP6ER1 and CarE1 are involved in nitenpyram resistance in Nilaparvata lugens

Kaikai Mao, Ruoheng Jin, Zhijie Ren, Junjie Zhang, Zhao Li, Shun He, Kangsheng Ma, Hu Wan, Jianhong Li

Summary: The study confirmed the relationship between nitenpyram resistance and overexpression of key metabolic enzyme genes CYP6ER1 and CarE1 in Nilaparvata lugens, and highlighted the importance of miRNAs in nitenpyram resistance. Global screening identified known and novel miRNAs associated with nitenpyram resistance, with two miRNAs (novel_85 and novel_191) targeting CYP6ER1 and CarE1, respectively, demonstrating their regulatory role in insecticide resistance. Modulating the expression of these miRNAs significantly altered nitenpyram susceptibility in N. lugens, suggesting their potential as new targets in insecticide resistance management.

Dynamics of Microbial Communities across the Life Stages of Nilaparvata lugens (Stal)

Zhijie Ren, Yunhua Zhang, Tingwei Cai, Kaikai Mao, Yao Xu, Chengyue Li, Shun He, Jianhong Li, Hu Wan

Summary: This study investigated the dynamic changes in bacteria and fungi in different developmental stages of the insect pest Nilaparvata lugens, revealing higher microbial diversity in eggs and mated adults compared to nymphs and unmated adults. It also found a strong correlation between fungal and bacterial alpha-diversity, suggesting a collaborative relationship between fungi and bacteria during the entire developmental period. The predominant bacterial and fungal taxa were identified, and the conservation of bacteria was greater than that of fungi during the transmission of the microbiota between developmental stages. The study highlights the potential value of further exploring microbial functions during different developmental stages and developing new pest management strategies.

Haoran Meng, Rong Huang, Hu Wan, Jianhong Li, Junkai Li, Xiaolei Zhang

Summary: Chilo suppressalis Walker is a devastating rice crop pest in China, and it has developed widespread resistance to many insecticides due to their intensive and extensive use. In this study, the susceptibility of 46 field populations of C. suppressalis to three insecticides was assessed. The results showed varying levels of resistance to the insecticides, and there was a significant correlation between the activity of detoxification enzymes and the toxicity of triazophos.

Microbiome variation correlates with the insecticide susceptibility in different geographic strains of a significant agricultural pest, Nilaparvata lugens

Yunhua Zhang, Tingwei Cai, Maojun Yuan, Zhao Li, Ruoheng Jin, Zhijie Ren, Yao Qin, Chang Yu, Yongfeng Cai, Runhang Shu, Shun He, Jianhong Li, Adam C. N. Wong, Hu Wan

Summary: Microbiome-mediated insecticide resistance in insect pests is influenced by host genotype, environmental factors, and the abundance of core bacterial symbionts. The expression of detoxifying genes in the host is correlated with the abundance of these symbionts and insecticide susceptibility levels. Moreover, environmental abiotic factors like temperature and precipitation can also predict symbiont abundance and insecticide susceptibility. These findings have implications for developing targeted microbial-based strategies to manage insecticide resistance in the field.

Insecticide resistance monitoring and diagnostics of resistance mechanisms in cotton-melon aphid, Aphis gossypii Glover in Central China

Dandan Shi, Tiantian Wang, Haixiang Lv, Xuchao Li, Hu Wan, Shun He, Hong You, Jianhong Li, Kangsheng Ma

Summary: This study determined the susceptibility of 15 field populations of Aphis gossypii in Central China to six chemical insecticides and explored the potential resistance mechanisms of these populations. The results showed that most populations have evolved varying degrees of resistance to imidacloprid, thiamethoxam, flonicamid, and pirimicarb. However, they remained susceptible to chlorpyrifos, and showed low to moderate levels of resistance to ?-cyhalothrin. Correlation analysis revealed that the enzyme activity of cytochrome P450 and carboxylesterase were associated with the resistance to certain insecticides. Additionally, multiple resistance-associated target site mutations were detected in the field populations. These findings provide important information for understanding insecticide resistance in China and designing effective strategies for managing A. gossypii.

Yongfeng Cai, Zhijie Ren, Chengyue Li, Tingwei Cai, Chang Yu, Qinghong Zeng, Shun He, Jianhong Li, Hu Wan

Summary: This study found that several fungicides have insecticidal activity against Nilaparvata lugens, with tebuconazole having the highest activity. Furthermore, tebuconazole significantly inhibited the expression of certain genes and affected the diversity and function of symbiotic fungi in N. lugens. These findings illustrate the insecticidal mechanism of tebuconazole and provide important rationale for delaying insecticide resistance.

Sublethal effects of acetamiprid and afidopyropen on Harmonia axyridis: insights from transcriptomics analysis

Liang Zhang, Haixiang Lv, Xuchao Li, Hu Wan, Shun He, Jianhong Li, Kangsheng Ma

Summary: In this study, the sublethal effects of acetamiprid and afidopyropen on Harmonia axyridis were evaluated through transcriptomics analysis. The results showed that sublethal concentrations of acetamiprid significantly reduced the reproductive capacity and lifespan of adult H. axyridis, while afidopyropen had limited effects. Additionally, both insecticides decreased H. axyridis predation. Transcriptome sequencing analysis revealed differential expression of detoxifying genes in response to acetamiprid and afidopyropen exposure. These findings highlight the importance of evaluating sublethal effects to protect and utilize natural enemies and guide pesticide use in the field.
