Kinetic modeling is the most suitable framework to describe the dynamic behavior of mammalian cell culture although its industrial application is still in its infancy. Herein, the authors reviewed mammalian bioprocess relevant kinetic models, and found that the simple unstructured-unsegregated approach utilizing empirical Monod-type kinetics based on limiting substrates and inhibitory metabolites is commonly used due to its traceability and simple formalism. Notably, the available kinetic models are typically small to moderate in size, and the development of large-scale models is severely hampered by the scarcity of kinetic data and limitations in current parameter estimation methods. The recent availability of abundant high-throughput multi-omics datasets from mammalian cell cultures have now paved the way to improve parameterization of kinetic models, and integrate regulatory, signaling, and product quality related intracellular events, as well as cellular metabolism within the modeling framework. Ultimately, the authors foresee that multi-scale modeling is the way forward in building predictive kinetic models of mammalian cell culture to advance biomanufacturing.

Insights into the Impact of Rosmarinic Acid on CHO Cell Culture Improvement through Transcriptomics Analysis

Summary: This study investigated the effects of the antioxidant rosmarinic acid (RA) on CHO cell cultures using transcriptomics analysis. The results showed that RA treatment modified gene expression and strongly affected the signaling pathways involved in cell survival and cell death. Furthermore, these signaling pathways continued to be up-regulated over time, indicating their importance in the cell culture process.

Summary: This study investigates the differences in growth, intestinal persistence, and postbiotic biosynthesis of six representative lactic acid bacteria (LAB) and their interactions with gut bacteria under different dietary regimes. The findings suggest that the attributes of probiotics are specific to both diet and species, and cannot be solely explained by genomics. Additionally, high-fat/low-carb diets may have detrimental effects on most LAB. The results highlight the importance of personalized probiotic design.

Mitigating biomass composition uncertainties in flux balance analysis using ensemble representations

Summary: The biomass equation in genome-scale metabolic models is used as the de facto objective function in flux balance analysis. However, the macromolecular composition of cells can change across different environmental conditions, raising concerns about the use of the same biomass equation in multiple conditions. Through qualitative and quantitative investigations, it was found that while macromolecular building blocks vary notably, the fundamental biomass monomer units are not appreciably different. Based on these findings, ensemble representations of the biomass equation were proposed to account for the natural variation of cellular constituents, leading to more accurate flux predictions.
