High Platelet-to-Lymphocyte Ratio Predicts Poor Prognosis and Clinicopathological Characteristics in Patients with Breast Cancer: A Meta-Analysis

Background. We aimed to evaluate the correlation of platelet-to-lymphocyte ratio (PLR) with prognosis and clinicopathological characteristics of breast cancer. Methods. The PubMed and Embase databases were searched. Hazard ratio (HR) with 95% confidence interval (CI) was used to summarize disease-free survival (DFS) and overall survival (OS). Odds ratio (OR) was used to summarize tumor clinicopathological characteristics. Results. High PLR was associated with poor DFS and OS (DFS: HR = 1.47, 95% CI = 1.16-1.85, and Tau(2) = 0.070; OS: HR = 1.88, 95% CI = 1.27-2.80, and Tau(2) = 0.192). A Galbraith plot indicated that the studies by Allan et al. and Cihan et al. contributed the heterogeneity of DFS and OS, respectively. There were significant differences in the incidence of high PLR between stage II-IV and stage I groups (OR = 1.86, 95% CI = 1.20-2.90, and Tau(2) < 0.001), between lymph node-positive and lymph node-negative groups (OR = 1.52, 95% CI = 1.22-1.91, and Tau(2) = 0.014), and between metastasis-positive and metastasis-negative groups (OR = 4.24, 95% CI = 2.73-6.59, and Tau(2) < 0.001). Conclusions. Our results indicated that PLR was associated with poor prognosis of breast cancer and adequately predicted clinicopathological characteristics.

Summary: YAP1 is highly upregulated in peritoneal carcinomatosis tumor cells, contributing to cancer stem cell properties and metastasis. Inhibition of YAP1 can significantly slow tumor growth and eliminate peritoneal carcinomatosis in preclinical models. Pharmacological inhibition of YAP1 can reduce cancer stem cell-like properties and suppress tumor growth, especially in combination with cytotoxic drugs in vivo models.

Increased Expression of LIPC Is Associated with Aggressive Phenotype of Borrmann Type 4 Gastric Cancer

Summary: High expression of LIPC is associated with poor clinical outcomes and plays an important role in regulating cell migration and invasion in Borrmann type 4 gastric cancer. High LIPC expression correlates with lymph node metastasis, advanced TNM stage, and poor overall survival in Borrmann type 4 gastric cancer patients. By reducing LIPC expression, cell invasion and migration are suppressed and specific gene expression is altered in Borrmann type 4 gastric cancer.

The Clinical Significance and Mechanisms of REG4 in Human Cancers

tRFTars: predicting the targets of tRNA-derived fragments

Summary: tRFs are small non-coding RNAs derived from specific tRNA cleavage events with key regulatory functions. Through identifying tRF-mRNA pairs and analyzing key features, a prediction model was constructed using genetic algorithm and support vector machine, which showed high accuracy in predicting tRF target genes. The user-friendly prediction tool, tRFTars, is available online for researchers to predict tRF targets in humans.

Survival landscape of different tumor regression grades and pathologic complete response in rectal cancer after neoadjuvant therapy based on reconstructed individual patient data

Summary: The study found that patients with rectal cancer achieving pathologic complete response (pCR) had significantly higher 10-year overall survival and 5-year disease-free survival compared to those in the non-pCR group. Improvement in tumor regression after neoadjuvant therapy was associated with increased overall survival and disease-free survival, and the pCR group had longer overall survival and disease-free survival than the non-pCR group. The results suggest a positive relationship between better tumor regressions and improved survival benefits, highlighting the importance of achieving pCR in the treatment of rectal cancer.

Single-cell sequencing of ascites fluid illustrates heterogeneity and therapy-induced evolution during gastric cancer peritoneal metastasis

Summary: Peritoneal metastasis is the leading cause of death for gastrointestinal cancers. By analyzing the single-cell transcriptomes of ascites cancer/immune cells from patients with/without gastric cancer peritoneal metastasis, this study reveals the increase of monocyte-like dendritic cells (DCs) during GCPM progression, the evolution of monocyte-like DCs and regulatory and proliferative T cells following therapy, and the high-plasticity GC clusters that exhibit a propensity to shift to a high-proliferative phenotype. The study also identifies autophagy-related genes as potential biomarkers of prognosis and demonstrates the role of autophagy inhibitors in inducing apoptosis in patient-derived organoids.
