Aim. The aim of the study was to evaluate the role of Interleukin-17 (IL-17), Interleukin-23 (IL-23), and transforming growth factor-beta (TGF-beta) in pregnancy complicated by placental insufficiency and in normal pregnancy. Material and Methods. The study comprised 34 patients with pregnancy complicated by fetal growth restriction (FGR) associated with preeclampsia (PE), as well as 35 healthy pregnant women. The concentrations of IL-17, IL-23, and TGF-beta in sera from maternal peripheral blood were determined by an immunoenzymatic assay. Results. There were higher concentrations of IL-17 in the study group when compared to the controls. In the group of patients with placental insufficiency, the levels of IL-17 positively correlated with systolic blood pressure (R = 0.42,P < 0.01). The study obtained comparable concentrations of IL-23 in both studied groups. The concentrations of TGF-beta were significantly lower in pregnancy complicated by the insufficiency of placenta when compared to the controls. Conclusions. It seems possible that the increased concentrations of IL-17 and the deficiency of TGF-.. in pregnancy complicated by FGR and PE can be responsible for the activation of inflammatory response observed in PE cases.

Insights of rheumatoid arthritis biomarkers

Bogdan Kolarz, Dominika Podgorska, Rafal Podgorski

Summary: Rheumatoid arthritis is a chronic autoimmune disease that affects joints and other organs, leading to disability and premature death if left untreated. Early recognition and treatment are crucial, and the disease can be classified into seropositive and seronegative based on specific antibodies. Ongoing research aims to understand disease mechanisms and identify markers for improved patient treatment.

Michal K. Zarobkiewicz, Ewelina Wawryk-Gawda, Wioleta Kowalska, Mariola Janiszewska, Agnieszka Bojarska-Junak

Summary: This paper reviews the role of gamma delta T cells in the pathogenesis and exacerbation of asthma, summarizing studies on humans and mouse models. It is found that gamma delta T cells play an important role in the pathogenesis of asthma, with different subsets likely to have opposite functions.

IL-15 Is Overexpressed in γδ T Cells and Correlates with Disease Severity in Relapsing-Remitting Multiple Sclerosis

Michal K. Zarobkiewicz, Wioleta Kowalska, Izabela Morawska, Pawel Halczuk, Konrad Rejdak, Agnieszka Bojarska-Junak

Summary: The study found a subset of IL-15+ gamma delta T cells in the blood of multiple sclerosis patients that correlates with disease severity. Further functional studies are needed to fully understand the importance of these cells in the pathogenesis of multiple sclerosis.

Prognostic Value of Tie2-Expressing Monocytes in Chronic Lymphocytic Leukemia Patients

Justyna Wos, Sylwia Chocholska, Wioleta Kowalska, Waldemar Tomczak, Agata Szymanska, Agnieszka Karczmarczyk, Agnieszka Szuster-Ciesielska, Agnieszka Wojciechowska, Agnieszka Bojarska-Junak

Summary: TEMs, characterized by the phenotype of CD14+CD16+Tie2+, have been identified as a new immunosuppressive force in tumors and are associated with unfavorable prognosis in CLL patients. Despite not being an independent predictor of survival, TEM can serve as an important complement to other prognostic indicators in CLL patients.

The Mysterious Actor-γδ T Lymphocytes in Chronic Lymphocytic Leukaemia (CLL)

Michal K. Zarobkiewicz, Agnieszka A. Bojarska-Junak

Summary: CLL is the most common leukemia among adults, and currently there is no cure. Despite the dysfunction of gamma delta T cells in CLL patients, they can still be used for immunotherapy.

Reduced Percentage of CD14dimCD16+SLAN+ Monocytes Producing TNF and IL-12 as an Immunological Sign of CLL Progression

Wioleta Kowalska, Michal Zarobkiewicz, Waldemar Tomczak, Justyna Wos, Izabela Morawska, Agnieszka Bojarska-Junak

Summary: Monocytes, particularly non-classical subpopulations, may play an important role in chronic lymphocytic leukemia (CLL) pathogenesis. This study assessed monocyte subsets divided based on SLAN and CD16 expression in CLL patients. The findings suggest an increased percentage of non-classical monocytes producing TNF and IL-12, while intermediate monocytes showed a higher percentage of cells synthesizing anti-inflammatory IL-10. The percentage of CD14(dim)CD16(+)SLAN(+) monocytes producing TNF and IL-12 decreased with CLL stage and correlated with prognostic factors. Notably, CLL patients requiring treatment had a lower percentage of CD14(dim)CD16(+)SLAN(+) monocytes producing TNF and IL-12, potentially indicating a beneficial effect of non-classical monocytes on anti-tumor response.

The lack of association between PADI4_94 or PADI4_104 polymorphisms and RF, ACPA and anti-PAD4 in patients with rheumatoid arthritis

Marek Ciesla, Bogdan Kolarz, Dorota Darmochwal-Kolarz

Summary: This study found no association between two polymorphisms of the PADI4 gene and levels of anti-PAD4, ACPA, and RF antibodies. Therefore, the genetic variants PADI4_94 and PADI4_104 may not be clinically relevant as prognostic factors in patients with established RA.

FCER1G Gene Hypomethylation in Patients with Rheumatoid Arthritis

Dominika Podgorska, Marek Ciesla, Bogdan Kolarz

Summary: This study analyzed the association between FCER1G gene methylation and RA activity. The results showed that RA patients have lower levels of methylation of the FCER1G gene, suggesting that FCER1G gene methylation may be a new potential epigenetic marker of RA that is independent of disease activity.

Sylwia Chocholska, Michal Zarobkiewicz, Agata Szymanska, Natalia Lehman, Justyna Wos, Agnieszka Bojarska-Junak

Summary: The aim of this study was to investigate the expression of miR-17 similar to 92 cluster members in CLL patients. The study provides new evidence regarding the heterogeneity of miR-17 similar to 92 cluster members' expression in CLL patients. High miR-20a expression can be considered an independent favorable prognostic factor.

Prognostic Potential of Galectin-9 mRNA Expression in Chronic Lymphocytic Leukemia

Agnieszka Bojarska-Junak, Wioleta Kowalska, Sylwia Chocholska, Agata Szymanska, Waldemar Tomczak, Michal Konrad Zarobkiewicz, Jacek Rolinski, Daniela De Totero, Paolo Giannoni

Summary: This study explores the potential role of Galectin-9 (Gal-9) as a novel prognostic biomarker in chronic lymphocytic leukemia (CLL). Through quantification of Gal-9 mRNA expression in purified B-lymphocytes of 100 CLL patients, the study reveals the upregulation of Gal-9 mRNA in malignant B-cells, which is closely associated with unfavorable prognostic markers and a shorter time to treatment. Furthermore, the study suggests that EBV coinfection may worsen prognosis in CLL by inducing upregulation of Gal-9 expression.
