Systematic analysis of nuclear localization of Autographa californica multiple nucleopolyhedrovirus proteins

Summary: This study systematically analyzed the nuclear localization of 154 ORFs encoded by AcMNPV, with 25 viral proteins found to be localized in the nucleus in the absence of virus infection. Upon virus infection, 11 proteins originally located in the cytoplasm were completely translocated into the nucleus, suggesting their nuclear import is facilitated by other viral or host proteins. Further experiments identified specific proteins that were imported into the nucleus with the assistance of other nuclear localization proteins.

Construction and Characterization of a Novel Bacmid AcBac-Syn Based on a Synthesized Baculovirus Genome

Summary: Researchers successfully modified a synthetic baculovirus, AcMNPV-WIV-Syn1, into a novel bacmid, AcBac-Syn, for use as a backbone for baculovirus system. By constructing a vector containing a LacZ:attTn7 and egfp cassette and recombining it with the AcMNPV-WIV-Syn1 genome in yeast, they were able to generate the bacmid AcBac-Syn. The resulting bacmid showed similar biological characteristics to the wild-type virus and could be easily used for foreign gene insertion and expression.

These findings provide alternative targets for the CRISPR antiviral system and offer new ideas for virus infection research and insect pest control.

AC81 Is a Putative Disulfide Isomerase Involved in Baculoviral Disulfide Bond Formation

Summary: The study identified that the baculovirus core gene ac81 likely encodes a disulfide isomerase which plays a crucial role in various functions of the baculovirus life cycle. This suggests that viral disulfide bond formation could be an ancient mechanism shared by insect-specific large DNA viruses like baculoviruses, nudiviruses, and hytrosaviruses encoding AC81 and P33 (sulfhydryl oxidase).
