Summary: The NUP98-PMX1 fusion gene, resulting from t(1;11)(q23;p15), is rare in acute myeloid leukemia (AML) patients. This report represents the first case of de novo AML carrying the NUP98-PMX1 fusion gene, contributing to a more comprehensive understanding of this rearrangement. Mechanistic studies are needed to fully elucidate the role of the NUP98-PMX1 fusion gene in leukemia pathogenesis.

Summary: This study analyzed the different fusion genes and chromosomal abnormalities in pediatric acute myeloid leukemia (AMkL) and evaluated the 5-year event-free survival (EFS), overall survival (OS), and relapse rate in these subtypes. The results showed that CBFA2T3::GLIS2 and KMT2A rearrangements were associated with worse prognosis compared to other AMkL, while RBM15::MRTFA or AMkl-MRC subtypes had better outcomes. NUP98 fusion genes were associated with poor prognosis in the AAML1031 trial.

Summary: A dieback was observed on three-year-old pot-grown plants of Cycas revoluta in Sicily (Italy), and Phytophthora species were identified as the causal agent. Among the three Phytophthora species isolated, P. pseudocryptogea was found to be the most virulent, while P. multivora was the least virulent. The pathogenicity tests confirmed that P. pseudocryptogea was responsible for the decline of C. revoluta.

Summary: We present a case of a 69-year-old female with human T-cell leukemia virus type 1 carrier who developed three hematological malignancies within a short period: diffuse large B-cell lymphoma (DLBCL), chronic myelomonocytic leukemia (CMMoL), and acute myeloid leukemia (AML). The AML cells showed features of acute promyelocytic leukemia (APL) but lacked the RAR alpha gene fusion, leading to initial diagnosis of APL-like leukemia (APLL). Genomic sequencing revealed a chromosomal rearrangement between KMT2A and ACTN4 in both CMMoL and APLL samples, suggesting their clonal relationship. Additionally, the APLL sample exhibited NRAS G12 mutation, suggesting its contribution to leukemic transformation. This report emphasizes the importance of sequencing analysis in understanding therapy-related leukemia.

Summary: This is a case report on a 27-year-old female with chronic myeloid leukemia (CML) who developed kinase-independent resistance and disease progression. The patient initially had chronic phase (CP) CML and acquired mutations in ASXL1 and RUNX1. Due to uncontrolled disease, she progressed to a chronic myelomonocytic leukemia-like (CMML) accelerated phase (AP) disease and obtained an IDH1 mutation. The disease progression was associated with the development of an inflammatory serositis, a phenomenon not typically observed in AP-CML.

Summary: This case report presents a female patient with ALK-rearranged metastatic lung adenocarcinoma who became pregnant while receiving alectinib treatment. With the collaborative effort of a multidisciplinary team, the patient continued the study drug throughout pregnancy and delivered a healthy baby girl. Both maternal and fetal parameters remained normal during the pregnancy, and the baby showed no developmental anomalies during the first 20 months of life. After 32 months from diagnosis, the mother is doing well and in partial remission.
