Endocrine-disrupting chemicals such as p,p'-dichlorodiphenyldichloroethylene (p,p'-DDE), are bioaccumulated in the adipose tissue (AT) and have been implicated in the obesity and diabetes epidemic. Thus, it is hypothesized that p,p'-DDE exposure could aggravate the harm of an obesogenic context. We explored the effects of 12 weeks exposure in male Wistar rats' metabolism and AT biology, assessing a range of metabolic, biochemical and histological parameters. p,p'-DDE-treatment exacerbated several of the metabolic syndrome-accompanying features induced by high-fat diet (HF), such as dyslipidaemia, glucose intolerance and hypertension. A transcriptome analysis comparing mesenteric visceral AT (vAT) of HF and HF/DDE groups revealed a decrease in expression of nervous system and tissue development-related genes, with special relevance for the neuropeptide galanin that also revealed DNA methylation changes at its promoter region. Additionally, we observed an increase in transcription of dipeptidylpeptidase 4, as well as a plasmatic increase of the pro-inflammatory cytokine IL-1 beta. Our results suggest that p,p'-DDE impairs vAT normal function and effectively decreases the dynamic response to energy surplus. We conclude that p,p'-DDE does not merely accumulate in fat, but may contribute significantly to the development of metabolic dysfunction and inflammation. Our findings reinforce their recognition as metabolism disrupting chemicals, even in non-obesogenic contexts.

Decreased adiponectin/leptin ratio relates to insulin resistance in adults with obesity

Ines Castela, Juliana Morais, Ines Barreiros-Mota, Marta P. Silvestre, Claudia Marques, Catarina Rodrigues, Shamila Ismael, Joao R. Araujo, Miguel angelo-Dias, Catarina Martins, Luis Miguel Borrego, Rosario Monteiro, Silvia Ribeiro Coutinho, Conceicao Calhau, Catia Martins, Ana Faria, Diogo Pestana, Diana Teixeira

Summary: This study found that the adiponectin/leptin ratio (AdipoQ/Lep) can be used as a biomarker of adipose tissue and metabolic function in adults with obesity and without diabetes. AdipoQ/Lep is associated with insulin resistance and correlates with obesity indices and anti-insulin hormone levels in the blood. These data highlight the role of adipocyte dysfunction in the pathogenesis of insulin resistance and suggest that AdipoQ/Lep may be a promising early marker of insulin resistance in adults with obesity.

White blood cells in a healthy adolescent population according to social and health characteristics

Isaac Barroso, Elisabete Ramos, Vanda Craveiro, Jao Tiago Guimaraes

Summary: This study examined the relationship between leukocytes and social and environmental determinants of health in adolescents. The results showed that adolescents with better socioeconomic conditions and those who participated in sports had lower levels of white blood cells, while those with chronic diseases or allergies had higher levels of white blood cells. In addition, an increase in body mass index and systemic inflammation was associated with higher levels of white blood cells.

Dose-response effect of a recreational team handball-based exercise programme on cardiometabolic health and physical fitness in inactive middle-aged-to-elderly males - a randomised controlled trial

Ivone Carneiro, Peter Krustrup, Carlo Castagna, Magni Mohr, Jose Magalhaes, Rita Pereira, Rute Santos, Sandra Martins, Joao Tiago Guimaraes, Eduardo Coelho, Susana Povoas

Summary: This study aimed to examine the dose-response of a recreational team handball exercise-based programme on cardiometabolic health and physical fitness in inactive middle-aged-to-elderly males without TH experience. The results showed that performing three 60-minute team handball exercise sessions per week can improve aerobic performance in middle-aged-to-elderly men, and it is more effective than training with a lower weekly frequency.

Fe3O4@Au Core-Shell Magnetic Nanoparticles for the Rapid Analysis of E. coli O157:H7 in an Electrochemical Immunoassay

Shayesteh Bazsefidpar, Maria Freitas, Clara R. Pereira, Gemma Gutierrez, Esther Serrano-Pertierra, Henri P. A. Nouws, Maria Matos, Cristina Delerue-Matos, Maria Carmen Blanco-Lopez

Summary: In this paper, a method for effectively controlling Escherichia coli (E. coli) O157:H7 in milk samples was developed. Monodisperse Fe3O4@Au magnetic nanoparticles were synthesized and used in an electrochemical sandwich-type magnetic immunoassay for rapid and accurate analysis. The assay showed a linear range from 20 to 2 x 10(6) CFU/mL, with a limit of detection of 20 CFU/mL for E. coli O157:H7. The selectivity of the assay was tested and the applicability was demonstrated by analyzing a commercial milk sample.

Contaminant Cocktails of High Concern in Honey: Challenges, QuEChERS Extraction and Levels

Mariana Lamas, Francisca Rodrigues, Maria Helena Amaral, Cristina Delerue-Matos, Virginia Cruz Fernandes

Summary: Environmental pollution is a significant issue in our society, and it is important to understand its consequences, including increased contaminant cocktails in the environment. The contamination of honeybees can lead to contamination of their products, such as honey. Controlling the quality and safety of honey, including evaluating contaminants and following legislation, is crucial.

Tracing the Path between Mushrooms and Alzheimer's Disease-A Literature Review

Ana Margarida Silva, Marco Preto, Clara Grosso, Monica Vieira, Cristina Delerue-Matos, Vitor Vasconcelos, Mariana Reis, Lillian Barros, Rosario Martins

Summary: Alzheimer's disease is a neurodegenerative disease that causes a decline in cognitive functions. It is the most common form of dementia and has no well-defined cause. Current treatments only manage to reduce symptoms, and there are only two drugs that can partially halt its progression. Natural compounds from mushrooms have been studied for their potential use in AD treatment due to their beneficial effects on various AD-related mechanisms such as enzyme inhibition, prevention of protein aggregation and neurotoxicity, and antioxidant and anti-inflammatory potential. While there have been studies linking mushrooms to neurodegenerative diseases, there is currently no publication summarizing the data specifically focused on AD. This review aims to fill that gap by compiling the potential anti-AD effects of mushroom extracts or compounds that target known AD-related mechanisms.

Stephanie L. Morais, Julia M. C. S. Magalhaes, Valentina F. Domingues, Cristina Delerue-Matos, Joilson Ramos-Jesus, Hygor Ferreira-Fernandes, Giovanny R. Pinto, Marlene Santos, M. Fatima Barroso

Summary: Cardiovascular diseases are a significant cause of death and illness. Warfarin is commonly used to treat these conditions, but its dosage variability can lead to complications such as hemorrhage or thromboembolism. The Cytochrome P450 (CYP) 2C9*3 genetic polymorphism contributes to this variability by modifying the enzymatic activity and plasmatic concentration of warfarin. Therefore, it is crucial to have a selective, rapid, low-cost, and real-time detection device before prescribing warfarin. In this study, a disposable electrochemical DNA-based biosensor capable of detecting CYP2C9*3 polymorphism was developed.
