Summary: Mitochondria play a crucial role in cellular processes beyond energy production, being involved in calcium homeostasis, ROS generation, cell death, thermogenesis, and biosynthesis. Glucocorticoids regulate mitochondrial gene expression and metabolism, with GR directly influencing mitochondrial functions including apoptosis and inflammation. Further studies are needed to fully understand the mechanisms of GR translocation and its impact on mitochondrial processes.

Summary: Deoxyelephantopin (DET), extracted from Elephantopus scaber L., shows significant inhibitory effects on colon cancer (CC) by inducing G2/M phase arrest, apoptosis, and DNA damage. DET reduces the expression of Bcl2 and miR-205, promoting CC cell apoptosis, and enhances the chemotherapeutic sensitivity of CC to 5-Fluorouracil (5FU). The study highlights the potential of DET as a potent drug for CC treatment, either alone or in combination with 5FU, and emphasizes the importance of the miR-205-Bcl2 axis in DET treatment.

Summary: SRPK1 is overexpressed in colon cancer and correlated with clinical stage and TNM classifications. High levels of SRPK1 are associated with poor prognosis. Mechanistically, SRPK1 enhances the anti-apoptosis ability of colon cancer cells under oxaliplatin treatment by promoting NF-κB pathway activation.

Summary: The E3 ligase SIAH1 is decreased in epithelial ovarian cancer (EOC) and negatively correlated with RPS3 levels. Overexpression of SIAH1 can suppress tumor cell growth, invasion, and cisplatin resistance by promoting RPS3 degradation and subsequent NF-kappa B inactivation.

Summary: Macrophages and microglia play critical roles in neuroinflammation, and the deletion of GRIP1 can reduce the severity of EAE. Interestingly, the loss of GRIP1 also blunted the therapeutic properties of IFN-beta while promoting the persistence of a homeostatic microglia signature. The results uncover the multifaceted function of type I IFN in the pathogenesis and therapy of MS/EAE, and reveal an unexpectedly permissive role of myeloid cell GRIP1 in neuroinflammation.

Summary: The study demonstrates that rutaecarpine (RUT) can inhibit the proliferation and migration of colorectal cancer cells and induce cell apoptosis by inactivating NF-kappa B/STAT3 signaling pathway. This suggests the potential clinical application of RUT for the treatment of colorectal cancer.

Summary: Levels of OGT and hyper-O-GlcNAcylation are associated with cancer pathogenesis, and combining OGT inhibitor OSMI-1 with TRAIL shows a synergistic effect in inducing apoptosis in cancer cells by increasing DR5 expression and activating ER stress response. This combination treatment inhibits NF-kappa B signaling, leading to enhanced TRAIL-induced apoptosis through caspase-8 activation in HCT116 cells.

Summary: The study demonstrates that TRAIL and Smac mimetics can induce non-canonical NF-kappa B and IFN signaling in certain breast cancer cell lines, emphasizing their potential therapeutic relevance. Phosphorylation of STAT1 is identified as a late event induced by TRAIL, independent of non-canonical NF-kappa B, and related to non-apoptotic role of caspase-8.

Summary: Pre-treatment with INI-43 significantly enhances cisplatin sensitivity in cervical cancer cells by stabilizing p53 and reducing nuclear import of NF kappa B. The combination of nuclear import inhibition and cisplatin shows synergistic effects in enhancing DNA damage and cell death in cervical cancer cells.

Summary: The study found that lower levels of FUT8 in osteosarcoma patients and cell lines are associated with the growth and progression of OS, with attenuated FUT8 expression leading to decreased levels of core-fucosylation of TNFRs, activating non-canonical NF-kappa B signaling pathway and reducing apoptosis in OS cells. FUT8 functions as a negative regulator of OS that enhances OS-cell apoptosis, suggesting a potential novel therapeutic strategy.
