Litchi is a tasty fruit that is commercially grown for food consumption and nutritional benefits in various parts of the world. Due to its biological activities, the fruit is becoming increasingly known and deserves attention not only for its edible part, the pulp, but also for its peel and seed that contain beneficial substances with antioxidant, cancer preventive, antimicrobial, and anti-inflammatory functions. Although literature demonstrates the biological activity of Litchi components in reducing tumor cell viability in in vitro or in vivo models, data about the biochemical mechanisms responsible for these effects are quite fragmentary. This review specifically describes, in a comprehensive analysis, the antitumor properties of the different parts of Litchi and highlights the main biochemical mechanisms involved.

Summary: The study investigates the biological activity and mechanism of action of the essential oil from Foeniculum vulgare subsp. piperitum fruits (FVPEO) in breast cancer cells. FVPEO exhibits strong anticancer effects by inhibiting cell growth and inducing apoptosis. The cytotoxic effect of FVPEO is associated with an increase in reactive oxygen species and stress-related proteins.

Summary: This study determined the phytochemical composition of different extracts of Helianthemum lippii and evaluated their anticancer, antimicrobial, and antibiofilm activities. The extracts showed cytotoxic activity against human MDA-MB231 and HCT116 cells and antimicrobial activity against certain bacteria and fungi. These extracts demonstrate interesting therapeutic potential.

The Histone Deacetylase Inhibitor ITF2357 (Givinostat) Targets Oncogenic BRAF in Melanoma Cells and Promotes a Switch from Pro-Survival Autophagy to Apoptosis

Summary: This study compared the effects of two HDAC inhibitors, ITF2357 and SAHA, in melanoma cells with the BRAF V600E oncogenic mutation. Results showed that ITF2357 was more effective than SAHA in reducing the viability of BRAF-mutated melanoma cells, and it also decreased the expression of oncogenic BRAF protein and induced a switch from autophagy to classic apoptosis.

Oncogenic BRAF and p53 Interplay in Melanoma Cells and the Effects of the HDAC Inhibitor ITF2357 (Givinostat)

Summary: We previously showed that the HDAC inhibitor ITF2357 targets oncogenic BRAF in melanoma cells. In this study, we found that oncogenic BRAF localizes to the nucleus of these cells and ITF2357 reduces both nuclear and cytosolic BRAF levels. We also observed an interaction between BRAF and oncogenic p53, and ITF2357 decreased levels of both proteins in certain melanoma cells.

A Deadly Liaison between Oxidative Injury and p53 Drives Methyl-Gallate-Induced Autophagy and Apoptosis in HCT116 Colon Cancer Cells

Summary: Methyl gallate (MG), a gallotannin found in plants, showed potential as an anti-tumor phytomolecule for colon cancer treatment. It reduced the viability of HCT116 colon cancer cells but had no effect on differentiated Caco-2 cells. MG induced early ROS generation and ER stress, leading to autophagy and apoptotic cell death. The increase in p53 played a crucial role in the mechanism, which was tightly linked to oxidative injury.

Seseli bocconei Guss. and S. tortuosum subsp. maritimum Guss. essential oils inhibit colon cancer cell viability

Summary: This study investigates the chemical compositions of essential oils extracted from different parts of two wild endemic species of Sicily, Seseli bocconei Guss. and Seseli tortuosum subsp. maritimum Guss. It is found that the main metabolite classes vary among the different parts of the plants. The stem essential oils of both plants exhibit strong cytotoxic effects against colon cancer cells, and germacrene D is identified as a promising molecule with anticancer properties.

Inhibition of MCL1 using A-1210477 BH3-mimetic promotes anoikis/apoptosis, impairs focal adhesions, reduces migration and invasiveness, and downregulates EMT and stemness markers in MDA-MB-231 cell line.

Summary: Inflammatory bowel disease (IBD) is a chronic inflammatory disorder that affects the gastrointestinal tract (GT) and is caused by genetic, microbial, and environmental factors. Dysbiosis, characterized by a decrease in beneficial bacteria and an increase in pathogenic species, is closely related to inflammation and oxidative stress. Dietary polyphenols, as natural antioxidants, can maintain redox equilibrium in the GT and prevent gut damage and inflammatory responses.
