Prognostic Stratification of Patients With Advanced Oral Cavity Squamous Cell Carcinoma

Purpose of Review Prognosis of advanced oral squamous cell carcinoma remains a challenge for clinicians despite progress in its diagnosis and treatment over the past decades. In this review, we assessed clinicopathological factors and potential biomarkers along with their prognostic relevance in an attempt to develop optimal treatment strategies for these patients. Recent Findings In addition to several pathologic factors that have been proposed to improve prognostic stratification and treatment planning in the eighth edition of the American Joint Committee staging manual on cancer, we reviewed some other imaging and clinicopathological parameters demonstrated to be closely associated with patient prognosis, along with the biomarkers related to novel target or immune therapy. Summary Evaluation of current literature regarding the prognostic stratification used in contemporary clinicopathological studies and progress in the development of targeted or immune therapy may help these patients benefit from tailored and personalized treatment and obtain better oncological results.

Summary: The study found that Taiwanese patients with resected oral cavity squamous cell carcinoma who underwent free flap reconstruction had a higher frequency of clear margins and a significant survival advantage compared to those who received local flaps. Factors such as higher age, advanced tumor staging, greater depth of tumor, and extranodal extension were associated with poorer prognosis for disease-specific survival and overall survival.

The impact of depression on survival of head and neck cancer patients: A population-based cohort study

Summary: This study found that depression is common among patients with head and neck cancer and that depression treatment has a significant impact on their survival outcomes. Patients diagnosed with depression before cancer and who did not receive psychiatric clinic visits after their cancer diagnosis had poorer survival rates. However, in advanced-stage patients, depression treatment may improve overall survival.

Development of a salivary autoantibody biomarker panel for diagnosis of oral cavity squamous cell carcinoma

Summary: Oral cavity squamous cell carcinoma (OSCC) is a common fatal illness, and early detection is urgently needed. We identified OSCC-associated autoantibodies using salivary samples and affinity mass spectrometry. Four autoantibodies were significantly increased in early-stage OSCC patients and can serve as potential biomarkers for early diagnosis.

Summary: This study analyzed the association between plasma matrix metalloproteinase-1 (MMP-1) levels and clinicopathological factors as well as survival outcomes in oral cavity squamous cell carcinoma (OSCC). The results showed that plasma MMP-1 levels were significantly associated with the severity and prognosis of OSCC.

Discovering the Clinical and Prognostic Role of Pan-Immune-Inflammation Values on Oral Cavity Squamous Cell Carcinoma

Summary: A newly introduced pan-immune-inflammation value (PIV) was not evaluated for its role in oral cavity squamous cell carcinoma (OSCC). In this study, the PIV was calculated with the following equation (neutrophil count x platelet count x monocyte count)/lymphocyte count from the results of the automated hematology analyzers in 853 OSCC patients from 2005 to 2017. Significant differences were observed for clinicopathological factors between higher and lower PIV patients, and higher PIV was associated with worse overall survival, disease-free survival, locoregional recurrence-free survival, and distant metastasis-free survival. Multivariate analyses demonstrated that PIV was an independent prognostic factor for overall and distant metastasis-free survival.

MFI2 upregulation promotes malignant progression through EGF/FAK signaling in oral cavity squamous cell carcinoma

Summary: By comparing DEGs in OSCC-TCGA patients with CNVs identified in OSCC-OncoScan dataset, we found 37 dysregulated candidate genes, 26 of which have been reported in HNSCC. Among 11 novel candidates, MFI2 was identified as the most significant prognostic molecular in OSCC. Knockdown of MFI2 reduced cell viability, migration, and invasion in OSCC cells through modulation of EGF/FAK signaling.

Summary: This study investigated the prognostic value of radiomics based on computed tomography (CT) for patients with laryngeal and hypopharyngeal cancer (LHC) during radiotherapy (RT). The results showed that the radiomic models based on peritumoral and intratumoral features during mid-RT had better predictive performance for overall survival (OS) than the models based on pre-RT intratumoral features. The mid-RT radiomic models were also found to be associated with treatment-related lymphopenia.
