KMT2A promotes melanoma cell growth by targeting hTERT signaling pathway

Melanoma is an aggressive cutaneous malignancy, illuminating the exact mechanisms and finding novel therapeutic targets are urgently needed. In this study, we identified KMT2A as a potential target, which promoted the growth of human melanoma cells. KMT2A knockdown significantly inhibited cell viability and cell migration and induced apoptosis, whereas KMT2A overexpression effectively promoted cell proliferation in various melanoma cell lines. Further study showed that KMT2A regulated melanoma cell growth by targeting the hTERT-dependent signal pathway. Knockdown of KMT2A markedly inhibited the promoter activity and expression of hTERT, and hTERT overexpression rescued the viability inhibition caused by KMT2A knockdown. Moreover, KMT2A knockdown suppressed tumorsphere formation and the expression of cancer stem cell markers, which was also reversed by hTERT overexpression. In addition, the results from a xenograft mouse model confirmed that KMT2A promoted melanoma growth via hTERT signaling. Finally, analyses of clinical samples demonstrated that the expression of KMT2A and hTERT were positively correlated in melanoma tumor tissues, and KMT2A high expression predicted poor prognosis in melanoma patients. Collectively, our results indicate that KMT2A promotes melanoma growth by activating the hTERT signaling, suggesting that the KMT2A/hTERT signaling pathway may be a potential therapeutic target for melanoma.

Rational engineering of a metalloprotease to enhance thermostability and activity

Summary: The rational design of enzymes with enhanced thermostability is efficient. This study successfully improved the thermostability and synthetic activity of a metalloprotease through a rational strategy, resulting in mutants with increased stability and enhanced Z-aspartame synthesis activity. Molecular dynamics simulation revealed that increased hydrogen bond interactions and a regional hydration shell were key factors in enhancing thermostability. This research provides valuable insights for enzyme engineering and has significant implications in industrial applications.

In-depth analysis of large-scale screening of WRKY members based on genome-wide identification

Summary: With high-throughput sequencing technology, a total of 74 WRKY genes were identified from the genome of Dendrobium officinale. Detailed analysis of gene structure, conserved motifs, and phylogenetic relationships revealed that these genes can be classified into three main groups and five subgroups. Chromosomal localization analysis suggested a possible genome duplication event. Expression analysis suggested the involvement of certain WRKY genes in the JA signaling pathway.

Pseudoprogression after advanced first-line endocrine therapy in metastatic breast cancer with bone metastasis: A case report

Summary: Approximately 75% of patients with advanced breast cancer develop bone metastasis, which significantly affects both the quality of life and the survival rate of patients. Accurate determination of the status of bone metastases is important for developing treatment strategies and the prognosis of the disease. This case report highlights the significance of distinguishing osteoblastic bone repair from real bone lesion progression in the clinical setting and provides new insights into strategies for the treatment of bone metastasis.

Study on effects of NH3 and/or H2 addition on the characteristics of soot formation and gas emissions in a laminar ethylene diffusion flame

Summary: The effects of NH3 and/or H2 addition on soot formation and gas emission in a laminar ethylene diffusion flame were studied. NH3 addition can suppress PAH and soot formation by both dilutive and chemical effects, while H2 addition chemically promotes soot formation. The soot formation of NH3-H2 blends is between flames doped with pure NH3 and H2.

Prediction of the transport, deposition, and absorption of multicomponent E-cigarette aerosols in a subject-specific mouth-to-G10 human respiratory system

Summary: Predicting the transport and deposition of e-cigarette aerosols in human respiratory systems is crucial for understanding the absorption of nicotine in the lung. This study used a computational fluid-particle dynamics (CFPD) method to model the aerosol transport dynamics and found that acidity levels in e-liquid can affect nicotine evaporation and delivery to the airways. The study also investigated the impact of e-liquid composition on droplet and vapor phase transport and absorption.

Effect of annealing on the properties of plasma-enhanced atomic layer deposition grown HfO2 coatings for ultraviolet laser applications

Summary: HfO2 monolayer coatings were prepared by plasma-enhanced atomic layer deposition (PEALD), and the effects of thermal annealing atmosphere and temperature on the properties of the coatings were investigated. The results showed that annealing in an oxygen atmosphere led to the decomposition of impurity compounds and the elimination of oxygen deficiency, resulting in lower absorption and higher laser-induced damage threshold (LIDT). The highest LIDT of 6.3 J/cm2 was obtained at an annealing temperature of 700°C, which was approximately three times higher than the as-deposited coating. This experimental result could be valuable for improving the LIDT of oxide coatings grown by PEALD or other similar methods.

Flavonoids: a review on biosynthesis and transportation mechanism in plants

Challenges still exist, but gaining a deeper understanding of these transport mechanisms holds great potential for various fields. This review aims to provide an overview of recent advancements in flavonoid transport mechanisms.

Differential involvement of WRKY genes in abiotic stress tolerance of Dendrobium huoshanense

Summary: This study identified 64 WRKY genes in the genome of Dendrobium huoshanense and revealed their involvement in responding to both biotic and abiotic stresses. The subcellular localization analysis showed that most of the WRKY genes were located in the nucleus. The evolutionary analysis provided insights into the origin and duplication events of the WRKY genes in Dendrobium species, and identified DhWRKY42 as a key regulator involved in the regulation of jasmonic-acid-responsive genes.

Three-Dimensional Imaging of Emulsion Separation through Liquid-Infused Membranes Using Confocal Laser Scanning Microscopy

Summary: Membrane technology is effective for treating emulsified oil-water mixtures, but fouling of the membrane reduces its capacity. Liquid-infused membranes (LIMs) show potential for resolving fouling issues, but their low permeate flux is a limitation. By using confocal laser scanning microscopy (CLSM) to analyze 3D images, we observe the coalescence of oil droplets and formation of oil channels in LIMs, which are influenced by factors such as oil concentration, infused liquid viscosity, and operating pressure. This study provides insights into the selective permeation of dispersed liquid phases through LIMs.

Genome sequencing-based transcriptomic analysis reveals novel genes in Peucedanum praeruptorum

Summary: This study identified bolting-related genes in Peucedanum praeruptorum through transcriptome and genome analysis, and found that these genes are involved in transcription regulation, defense response, and carbohydrate metabolism. Genes related to lignin polymerization and phosphatidylinositol signaling system were also identified. These findings provide new insights for further research on the growth and medicinal properties of Peucedanum praeruptorum.

Multi-scale characterization of organic matter pore space in deep marine shale combined with mathematical morphology

The study focused on the impact of heterogeneity and deposition on the organic matter pore space, and obtained key findings.

Functional and kinetics of two efficient phenylalanine ammonia lyase from Pyrus bretschneideri

Photosynthetic machinery and antioxidant enzymes system regulation confers cadmium stress tolerance to tomato seedlings pretreated with melatonin

Summary: This study found that the application of melatonin significantly enhanced various aspects of tomato plant growth and reduced the damage caused by cadmium stress. By increasing the activity of antioxidant enzymes and decreasing the levels of oxidative stress markers, melatonin improved the resistance of tomato plants to cadmium stress.
