Dimethyleacetamide improves the cryosurvivability of Indian red jungle fowl (Gallus gallus murghi) sperm

It was hypothesized that dimethyleacetamide (DMA) can be used as an alternate to glycerol for cryopreservation of Indian red jungle fowl semen. Four concentrations of DMA (4%, 6%, 8% and 10%) in extender were compared with previously optimized cryopreservation protocol based on 20% glycerol (control) for Indian red jungle fowl. Sperm motility, plasma membrane integrity, viability, and acrosome integrity were assessed at the stage of post-dilution, cooling, equilibration, and freeze-thawing. The whole experiment was repeated/replicated for five times independently. Sperm motility, plasma membrane integrity, viability and acrosome integrity were recorded highest (P < 0.05) at post-dilution, cooling, equilibration, and freeze-thawing in extender having 6% DMA compared to control and other experimental extenders. The highest (P < 0.05) recovery rates of all aforementioned parameters were also recorded in extender having 6% DMA; thus, 6% DMA was further compared with control (20% glycerol) for fertility after artificial insemination. Eggs were collected for five days after artificial insemination with semen cryopreserved in extender containing 6% DMA and control. The higher no. of fertilized eggs, fertility, no. of hatched eggs, hatch (%) and hatchability were recorded with semen cryopreserved in extender having 6% DMA compared to control. It is concluded that 6% DMA maintained higher post-thaw quality and fertility of Indian red jungle fowl semen and is a better replacement of glycerol. (C) 2017 Elsevier Inc. All rights reserved.

Summary: This study compared the effects of two cryoprotectants, DMA and DMSO, on the frozen-thawed sperm characteristics of black-footed penguins and gentoo penguins. The results showed that 8% DMSO was more effective for black-footed penguin sperm, while there were no significant differences in gentoo penguin samples. Additionally, black-footed penguin sperm was more sensitive to the freezing-thawing process compared to gentoo penguin sperm.

Summary: The study aimed to investigate the influence of osmotic changes on the location of aquaporins (AQPs) in spermatozoa during freeze-thawing process. Individual variability in terms of freezeability was observed among rams and their ejaculates. AQP3 expression in the mid- and principal piece of spermatozoa was found to be higher in frozen-thawed samples compared to fresh specimens in the good freezability group, but such differences were not detected in the poor freezability group. AQP3 expression is associated with sperm cryotolerance and can be used as a biomarker.

Review: Sperm cryopreservation in wild small ruminants: morphometric, endocrine and molecular basis of cryoresistance

Summary: Reproductive technologies can help protect wild ruminant species from extinction and increase the number of animals. Efforts have focused on preserving sperm, oocytes, and embryos. Cryopreservation of sperm is the most commonly used and cheapest method for germplasm storage. New reproductive biotechnologies, such as ultra-rapid freezing techniques, have been developed. However, the success of artificial insemination using frozen-thawed sperm is not always satisfactory in wild small ruminants. Epididymal sperm shows higher cryoresistance than ejaculated sperm, and the seasonal variation in testosterone and prolactin levels is involved in sperm cryoresistance.

This improvement is attributed to the conversion of lactate to pyruvate and increased NAD(+) enhancing glycolysis efficiency.

Differential transcript usage across mammalian oocytes at the germinal vesicle and metaphase II stages

Summary: Ongoing progress in mRNA-Sequencing technologies has contributed to the refinement of assisted reproductive technologies. This study utilized mRNA-Seq datasets of various mammalian species to analyze differential transcript usage (DTU) and identified ten cross-species DTU genes. Gene Ontology (GO) term enrichment analysis revealed the involvement of these genes in RNA and cell-cycle control mechanisms, suggesting their importance during oocyte maturation. Further exploration of these genes' transcript isoforms may lead to the discovery of novel markers for reproductive technologies.

Effects of Nobiletin supplementation on the freezing diluent on porcine sperm cryo-survival and subsequent in vitro embryo development

Summary: This study found that Nobiletin (NOB) can promote the cryosurvival and embryo development of porcine sperm after in vitro fertilization (IVF). The addition of NOB improves sperm motility, acrosome integrity, viability, and mitochondrial membrane potential, reduces lipid peroxidation, and increases cleavage and blastocyst formation rates. Furthermore, NOB treatment upregulates the expression of genes related to pluripotency and cell differentiation in the resulting embryos.

Fluorochloridone induces mitochondrial dysfunction and apoptosis in primary goat Sertoli cells

Summary: This study investigated the cytotoxicity and toxicological mechanisms of fluorochloridone (FLC) on cultured goat Sertoli cells. The results showed that FLC exposure induced oxidative stress, promoted apoptosis and autophagy, inhibited lysosomal biogenesis, and blocked autophagic flux in goat Sertoli cells. FLC also caused mitochondrial dysfunction and disturbed mitophagy, leading to cell apoptosis. The antioxidant N-acetyl-L-cysteine (NAC) reduced FLC-induced ROS accumulation and reversed the disorder of autophagy levels.
