Recent clinical studies report antipsychotics as a better option to augment the action of antidepressants in treatment resistant cases. However, the proper mechanisms underlying the antidepressant effect of anti psychotics is still not clear. Indolamine 2, 3 dioxygenase (IDO) pathway is considered to be an important pathway in pro-inflammatory cytokine associated stress-induced depression. The present study investigated the antidepressant effect of venlafaxine, olanzapine and their combinations in chronic forced-swim stress-induced depression in mice. In addition, the role of pro-inflammatory cytokines and IDO-mediated pathway was investigated. Swiss albino mice were subjected to chronic forced-swim stress and evaluation for antidepressant-like activity was performed on 7th, 14th and 21st day following which serum levels of pro-inflammatory cytokines (IL-1 beta and IL-6 levels) and the levels of hippocampal kynurenine (KYN), tryptophan (TRP) and serotonin (5HT) were estimated. The combination exhibited augmentation of antidepressant-like activity of venlafaxine by olanzapine in chronic forced-swim stress model. Further, it reversed the enhanced serum IL-1 beta and IL-6 and reverted the increased activity of IDO as measured by ratio of hippocampal KYN/TRP and 5HT/TRP in stressed mice. Augmentation of antidepressant effect of venlafaxine by olanzapine is thus mediated by normalising the shift from KYN to TRP pathway in chronic forced swim induced stressed mice.

Development, optimisation and evaluation of chitosan nanoparticles of alendronate against Alzheimer's disease in intracerebroventricular streptozotocin model for brain delivery

Summary: The study aimed to develop ALN-loaded chitosan nanoparticles for brain delivery via the intranasal route. The CS-ALN-NPs showed reduced peripheral side effects, direct release of ALN to the brain, and controlled release properties. The optimized NPs demonstrated a smaller particle size within the 200nm range and sustained drug release, making them a promising brain-targeted delivery system for Alzheimer's disease treatment.

Summary: Melatonin and its analogues are considered to have antiseizure effects, but further research is needed to investigate their mechanisms of action and potential implications in the treatment of epilepsy. Evaluating melatonergic signaling pathways and mechanisms may lead to significant advancements in the field of epileptology in the future.

Summary: Liposomes-based bone-targeted delivery of therapeutics shows promise for treating osteoporosis effectively. However, despite extensive in vitro and in vivo research on liposomes, significant progress in their clinical translation has not been achieved. Successful translation from bench to bedside requires interdisciplinary collaboration between preclinical and clinical experts at different stages of liposomes development.

Summary: Diabetes and osteoporosis pose significant burdens on quality of life and healthcare costs. Recent research has focused on the impact of diabetes on bone metabolism, as well as the effects of metformin on bone health in individuals with type 2 diabetes. While studies have shown beneficial effects of metformin on bone microarchitecture, bone mineral density, and fracture risk, there are limitations in animal and clinical studies that warrant further investigation.
