Summary: This study aimed to explore the barriers and facilitators to the uptake of COVID-19, influenza, and pneumococcal vaccines in immunosuppressed adults with immune-mediated inflammatory diseases (IMIDs). The results showed that key facilitators to all vaccines included higher perceived infection risk and belief in the benefits of vaccination, while key barriers included concerns about triggering IMID flare and active IMID. Specific facilitators for COVID-19 vaccines included media focus, high incidence, mass-vaccination program with visible impact, social responsibility, and confirmation by healthcare professionals. Awareness of eligibility, direct invitation, and clear recommendation from trusted healthcare professionals were identified as key facilitators for influenza/pneumococcal vaccines, while lack of awareness of eligibility, no direct invitation or recommendation, low perceived infection risk, and lack of perceived benefit were identified as key barriers.

Summary: This study investigated the barriers and facilitators to COVID-19, influenza, and pneumococcal vaccine uptake in immunosuppressed adults with immune-mediated inflammatory diseases. The results showed that factors influencing vaccination included perceived infection risk, disease activity, media focus, healthcare professionals' recommendation, and eligibility awareness. It is important to address vaccine benefits and safety for IMID patients in clinical practice, improve public health messaging, and provide direct invitations to optimize vaccine uptake.

Summary: This study selected culturing conditions for Rba. capsulatus cells with minimal capsule and demonstrated their effectiveness in transferring exogenous DNA through electroporation, compared with cells cultivated on other conventionally used media. It was found that the survival rate of the culture after electroporation of plasmid DNA into Rba. capsulatus cells was unstable, requiring optimization of obtaining electrocompetent cells and careful selection of electric-shock treatment mode. Additionally, the study demonstrated for the first time the possibility of using in vitro methylation with a cell-free extract of Rba. capsulatus as an approach to protect plasmid DNA from destruction by the restriction modification system and transform the treated DNA into competent cells through electroporation.
