Summary: This study evaluated the impact of rhein, an anthraquinone compound found in Chinese medicine, on the lysosomal compartment and its indirect influence on cell apoptosis. The results showed that high concentrations of rhein caused oxidative stress and changes in lysosomal membrane permeability, leading to cell apoptosis. In addition, the use of an autophagy inhibitor, chloroquine, enhanced the cytotoxicity of rhein.

Summary: Mitochondrial dysfunction and failure of mitophagy in familial Alzheimer's disease (fAD) are associated with the accumulation of amyloid precursor protein-derived C-terminal fragments (APP-CTFs). This study demonstrates that the accumulation of APP-CTFs within mitochondrial domains is both a cause and consequence of mitochondrial dysfunction, leading to impaired mitophagy function.

Summary: In this study, the mRNA level of YME1L1 was knocked down in pig embryos and the expression patterns of YME1L1 and related proteins were examined. It was found that YME1L1 is localized in the punctate structures of mitochondria and highly expressed from the 4-cell stage. Knocking down YME1L1 led to decreased blastocyst rate and quality, mitochondrial fragmentation, excessive ROS production, lower mitochondrial membrane potential and ATP levels. Moreover, YME1L1 knockdown induced OPA1 cleavage and cytochrome c release. These findings demonstrate that YME1L1 is crucial for regulating mitochondrial fission, function, and apoptosis during porcine embryo preimplantation development.

Summary: The regulation of the cell division cycle is important for maintaining stable genome and tissue homeostasis. Dysregulation can lead to genomic instability and tissue abnormalities, as shown by upregulation of Cyclin E in the accessory gland of Drosophila melanogaster. Upregulation of Cyclin E causes endoreplication, DNA damage, and mitochondrial defects, resulting in tissue dysplasia.

Summary: Three novel cyclometalated iridium(III) compounds were synthesized and evaluated for their anti-cancer properties. Among them, Ir1 showed high cytotoxicity against HeLa cells and triggered apoptosis by reducing mitochondrial membrane potential and depleting ATP levels.

Summary: Vanadocene dichloride (VDC), a vanadium containing metallocene dihalide, shows promising anticancer activity by inducing apoptotic cell death in mammalian cancer cells and disrupting microtubule assembly dynamics to inhibit cell proliferation. VDC also inhibits the ATPase activity of the mitotic kinesin Eg5, suggesting a potential mechanism for its antimitotic activity.

Summary: The study aimed to investigate whether NaAsO2 might induce mitochondrial damage and apoptosis of cervical cancer cells through NF-kappa B/miR-21/GCLC induced oxidative stress, and play the anti-tumor role of arsenic as a potential drug for the treatment of cervical cancer. Results showed that NaAsO2 could lead to mitochondrial apoptosis of cervical cancer cells, with involvement of NF-kappa B and miR-21.
