Effect of elicitors on xanthone accumulation and biomass production in hairy root cultures of Gentiana dinarica

The effect of abiotic (salicylic acid, jasmonic acid and methyl jasmonate) and biotic (chitosan and yeast extract) elicitors on the growth and xanthone accumulation in two hairy root clones of Gentiana dinarica Beck. was studied. The obtained results showed that clone 3 was more responsive to elicitor treatment than clone D. The production of dominant xanthone norswertianin-1-O-primeveroside was not significantly affected by either of the abiotic elicitor tested but was stimulated with chitosan treatment. The highest concentrations of all elicitors strongly increased the content of xanthone aglycone norswertianin, but simultaneously reducing the production of its glycoside norswertianin-1-O-primeveroside. The most efficient in enhancing norswertianin production was a 7-day treatment with salicylic acid (200 mu M) and chitosan (50 mg l(- 1)), which yielded a 7.7- and a 24-fold increase in norswertianin content, respectively. In addition, treatment with biotic elicitors caused the occurrence of new xanthone compounds that were not detected in other samples. Free radical scavenging activity of xanthones was carried out by DPPH assay, and norswertianin showed the strongest activity.

Summary: Natural populations of Gentiana asclepiadea were analyzed using HPLC to determine the contents of six secondary metabolites, showing clear separation between populations based on differences in compound content and origin. Bulb Dormancy In Vitro-Fritillaria meleagris: Initiation, Release and Physiological Parameters

Summary: Ornamental geophytes face challenges with slow development and ineffective propagation methods, leading to the development of micropropagation techniques to improve multiplication rate and shorten regeneration periods. Dormancy is a key limitation in vitro multiplication, as bulbs require overcoming dormancy for sprouting. Understanding dormancy processes can help accelerate breeding and achieve economic profitability in horticulture. This review specifically focuses on recent findings in bulb dormancy initiation and release in fritillaries, emphasizing the effects of plant growth regulators and low-temperature pretreatment on dormancy release and antioxidative enzyme activity induction in vitro.

Summary: In vitro shoot culture of the endangered medicinal plant Gentianella lutescens was established and different treatments were found to have an impact on shoot growth and xanthone production. Controlling the concentration of sucrose, sorbitol, and elicitors in the culture medium can significantly affect the accumulation of xanthones in shoot cultures.

Xanthones Production in Gentiana dinarica Beck Hairy Root Cultures Grown in Simple Bioreactors

Summary: The hairy root clones of Gentiana dinarica cultivated in TIS RITA(R) and BCB with 1/2 MS medium and 4% sucrose showed higher dry matter content and xanthones production compared to those in EF. Among the clones, clone cl-B displayed superior growth parameters in BCB and TIS RITA(R), with the highest production of nor-1-O-prim and norswertianin. The optimized conditions in highly aerated TIS RITA(R) and BCB systems contribute to the development of bioreactor technology for large scale commercial production of xanthones nor-1-O-prim and norswertianin.

Summary: This study investigated the VOC emissions of Clinopodium pulegium, an endangered species endemic to the Southern Carpathians. It was found that the presence and concentration of carbon sources influenced the VOC emissions and characteristics of leaf glandular trichomes, while the qualitative composition of the volatiles was only slightly affected.

The Effects of Different Auxin-Cytokinin Combinations on Morphogenesis of Fritillaria meleagris Using Bulb Scale Sections In Vitro

Summary: This study investigated the effects of various auxin-cytokinin combinations on different morphogenetic pathways in bulb scale culture of Fritillaria meleagris. Low concentrations of plant growth regulators were found to be effective for shoot multiplication, with up to 5.5 shoots per explant. The combination of 2,4-D and low BAP concentration produced the highest number of bulbs, while PGR-free medium was effective in somatic embryo formation.
