Summary: Extracellular vesicles (EVs) have emerged as a promising source of biomarkers, but current diagnostic methods face challenges. A highly sensitive double digital assay is developed, allowing for absolute quantification of single molecules from individual EVs. The technology successfully separates and detects single protein molecules from single EVs. The system is applicable for evaluating important cancer biomarkers and sheds light on EV heterogeneity and discovery of new EV subtype biomarkers.
