Summary: Fifteen undescribed diterpenoid alkaloids and seven known analogs were isolated from Aconitum tanguticum. Their structures were determined using spectroscopic methods. The compounds exhibited inhibitory effects on the secretion of NO and TNF-alpha in LPS-treated RAW 264.7 cells, with IC50 values ranging from 67.56 mu M to 683.436 mu M.

Summary: This study provides a comprehensive view of the A. carmichaelii transcriptome by using a combination of Pacbio isoform sequencing and RNA sequencing, identifying 19 TPSs and five alternative splicing isoforms belonging to TPS-b, TPS-c, and TPS-e/f subfamilies. In vitro enzyme reaction analysis functionally identified two sesqui-TPSs and twelve diTPSs. The data clarified ent-CPP as the sole precursor to all DAs biosynthesis, with multiple genes responsible for different types of DAs biosynthesis in A. carmichaelii, laying the foundation for further exploration of the C-20-DAs biosynthetic pathway in the Aconitum species.

Summary: This study provides evidence for the traditionally medicinal use of Aconitum heterophyllum in treating malaria. The chloroform fraction of the roots was found to have the highest anti-plasmodial activity, attributed to the presence of the compound 2-O-cinnamoyl hetisine. Additionally, a new UHPLC-DAD analytical method was developed for quantification of marker compounds and quality assessment of this medicinal plant.

Summary: This study identified four important markers in Aconitum carmichaelii and 15 markers to differentiate it from adulterant species. Mesaconitine and hypaconitine were found to be the principal toxins in fuzi, while fuziline and benzoylmesaconine were associated with its medicinal properties. The detection of additional marker compounds in other Aconitum species can be useful in identifying adulteration.

ADAs have low toxicity, are widely distributed in the body, and possess various pharmacological activities.

Summary: In this study, a new denudatine-type C-20-diterpenoid alkaloid named arcutisine was isolated from the roots of Aconitum fischeri var. arcuatum, along with 24 known compounds. The isolation of compounds 1 and 2 from natural sources for the first time, and the weak anti-inflammatory activity of compounds 7 and 10 at a concentration of 40 μM were notable findings.

Summary: Two unprecedented aconitine-type C19-diterpenoid alkaloids, named refractines A-B (1-2), were isolated from the roots of Aconitum refractum. In addition, 12 known compounds (3-14) were also identified. The structures of these compounds were determined through extensive spectroscopic analyses. The inhibitory activities of all compounds against NO production in LPS induced RAW 264.7 macrophages were tested, and compounds 10 and 14 showed slight inhibition with rates of 29.4% and 22.1% at a concentration of 30 μM, respectively.
