Summary: This study aimed to identify quality markers (Q-markers) that could differentiate the efficacy of promoting blood circulation and removing blood stasis of Vaccaria segetalis (VS) and wine-steamed Vaccaria segetalis (WVS), and to establish quality standards for WVS. The results showed that the efficacy of WVS was more prominent than VS, and five components were identified as effective components of WVS for promoting blood circulation and removing blood stasis. Four active ingredients were screened out as Q-markers to distinguish between VS and WVS. The content determination showed that the total contents of the Q-markers in WVS from different origins ranged from 0.478% to 0.716%.

Summary: This study investigated the therapeutic effects and active chemical components of prepared rhubarb on noxious heat blood stasis syndrome (NHBS) using metabolomics. The results showed that prepared rhubarb could regulate the changes of endogenous metabolites caused by NHBS and restore the disturbances in multiple metabolic pathways. Furthermore, rhein and its metabolites were identified as important active compounds in prepared rhubarb for its biomedical activities.

Summary: Zhuling Jianpi Capsule is a traditional Chinese medicinal formula used to treat symptoms of inflammatory bowel disease. This study evaluated the therapeutic effects of Zhuling on experimental ulcerative colitis in mice and identified its chemical constituents and metabolomics using advanced techniques. The results showed that Zhuling alleviated colitis symptoms, suppressed pro-inflammatory cytokines, and improved intestinal barrier function. The analysis also identified potential active compounds responsible for the therapeutic effects.

Summary: The impurity profile of Compound Glycyrrhizin Tablet (CGT) was investigated, and a novel strategy for characterizing saponin-related impurities was proposed. A total of 41 saponin-related impurities were identified in CGTs, and differences in impurity profiles among CGTs from different manufacturers were found. These findings provide technological support for evaluating saponin-related impurities and lay a solid foundation for developing strategies to improve product quality in the future.

Summary: In this study, the representative components of Sang Xing decoction (SXD) were characterized using liquid chromatography-tandem mass spectrometry (LC-MS). The key targets, signaling pathways, and metabolic pathways associated with SXD in the treatment of acute bronchitis were identified via network prediction and metabolomics. In vivo experiments showed that SXD may maintain the production of inflammatory factors by regulating the PI3K/Akt/NF-kappa B signaling pathway, improving the metabolism of glutamine and glutamate to reduce oxidative stress, and inhibiting apoptosis. Simultaneously, the possibility of using SXD as an adjuvant drug for COVID-19 treatment was also revealed.

Summary: Recent studies have shown that dysregulation of carboxyl submetabolome homeostasis is closely related to the occurrence and development of hepatocellular carcinoma (HCC). This study established a convenient and efficient workflow for absolute quantification of carboxyl submetabolome, which was applied to monitor carboxyl metabolic profile in human plasma of the whole progressive course of HCC. Biomarkers of health-hepatitis-cirrhosis-HCC were screened out and a diagnostic model was established. The relevancy between carboxyl submetabolome disorders and the pathogenesis of HCC was also investigated.

Summary: This study examines the differences in the clinical application scope of Chrysanthemum morifolium flower (CMF) and Chrysanthemum indicum flower (CIF), two herbs with similar origin. The study employs a combination of network pharmacology, molecular pharmacology, and metabolomics to investigate the similarities and differences in chemical components, efficacy, and mechanisms of action of the two herbs. The results show that while CMF and CIF have similar effects in lowering blood pressure, resistance to tissue damage, and improving dyslipidemia, they exhibit distinct effects in regulating inflammatory response, signaling pathways, lipid anabolism, and metabolic abnormalities. The study suggests that the comparable efficacies of CMF and CIF, despite their distinct mechanisms of action, may be attributed to the integration and counteraction of their different regulating capabilities.

Summary: The lack of adequate lipid reference standards is a major challenge for accurate quantitative analysis in lipidomics. Through the discovery of regularity and predictability in chromatography-mass spectrometry behaviors of lipids, a protocol and strategy were developed to calculate correlations between compound structures and analytical parameters within each lipid subclass. This allowed for the prediction of relevant LC-tandem-MS parameters for unknown lipids, and the establishment and validation of a quantification parameter database for 4081 lipids. This methodology was applied to monitor lipid metabolism changes in the plasma of health-hepatitis-cirrhosis-hepatocellular carcinoma (HCC) progression, and resulted in the identification of lipid biomarkers for distinguishing between health and different diseases, as well as different stages of disease.

Summary: Hepatocellular carcinoma (HCC) is the terminal stage of chronic liver diseases, and chronic inflammation may contribute to its formation. Dysregulation of bile acid homeostasis in the enterohepatic circulation is a key factor in the development of HCC. This study used a rat model to reproduce HCC and monitored the bile acid profile in plasma, liver, and intestine. Differences in bile acid levels, including potential biomarkers for early diagnosis of HCC, were identified. The study also identified a key enzyme, bile acid-CoA:amino acid N-acyltransferase (BAAT), involved in bile acid synthesis and the inflammatory-cancer transformation process. This research provides insights into the role of bile acid metabolism in the liver-gut axis during the inflammation-cancer transformation process and has implications for HCC diagnosis, prevention, and treatment.

Summary: The lungs and large intestine can regulate inflammation and immunity through the lung-gut axis. The transportation of gut microbiota and metabolites is an important communication channel. During the transition from ulcerative colitis (UC) to colorectal cancer (CRC), the composition of gut microbiota and metabolites related to inflammation changes significantly, and corresponding inflammatory changes also occur in lung tissues. By detecting the changes in pulmonary metabolites and microbiota, key markers related to lung and gut diseases can be identified.
