Type 1 iodothyronine deiodinase (DIO1) regulates peripheral metabolism of thyroid hormones that control cellular proliferation, differentiation and metabolism. The significance of DIO1 in cancer is unknown. In this study we hypothesized that diminished expression of DIOl, observed in renal cancer, contributes to the carcinogenic process in the kidney. Here, we demonstrate that ectopic expression of DIO1 in renal cancer cells changes the expression of genes controlling cell cycle, including cyclin El and E2F5, and results in inhibition of proliferation. The expression of genes encoding collagens (COL1A1, COL4A2, COL5A1), integrins (1TGA4, 1TGA5, 1TGB3) and transforming growth factor-beta-induced (TGFB1) is significantly altered in renal cancer cells with induced expression of DI01. Finally, we show that overexpression of DIO1 inhibits migration of renal cancer cells. In conclusion, we demonstrate for the first time that loss of DIO1 contributes to renal carcinogenesis and that its induced expression protects cells against cancerous proliferation and migration. TGF-β1 affects the renal cancer miRNome and regulates tumor cells proliferation

Karolina Hanusek, Beata Rybicka, Piotr Poplawski, Anna Adamiok-Ostrowska, Katarzyna Gluchowska, Agnieszka Piekielko-Witkowska, Joanna Boguslawska

Summary: TGF-beta 1 has been found to regulate the expression of multiple microRNAs involved in renal cell carcinoma (RCC), forming a complex network. It controls RCC cell proliferation and modifies the expression of oncogenic regulators, including ETS1. Additionally, TGF-beta 1 regulates the expression of several miRNAs and reciprocally modifies the expression of genes encoding TGF-beta 1 receptors and SMADs, suggesting a novel feedback mechanism mediated through miRNAs.

Nucleolar Proteins and Non-Coding RNAs: Roles in Renal Cancer

Piotr Poplawski, Joanna Boguslawska, Karolina Hanusek, Agnieszka Piekielko-Witkowska

Summary: Renal cell cancer, especially clear cell renal cell carcinoma (ccRCC), is known for its high aggressiveness and poor prognosis for patients. The nucleoli, as crucial substructures of nuclei, play a significant role in controlling cellular functioning and homeostasis through multiple non-coding RNAs, such as snoRNAs. Dysregulation of snoRNAs, including SNHG1, SNHG4, and SNHG12, in ccRCC tumors contributes to the aberrant expression of oncogenes and tumor suppressors, directly impacting ccRCC development and progression.

Summary: piRNAs are a class of small non-coding RNAs that regulate transposon and gene expression through mechanisms such as DNA methylation, histone modifications, and mRNA degradation. Recent studies suggest that piRNAs could serve as potential biomarkers for early cancer detection and therapeutic interventions, playing a role in the progression of prostate cancer.

Adenoid hypertrophy in children: a narrative review of pathogenesis and clinical relevance

Artur Niedzielski, Lechoslaw Pawel Chmielik, Grazyna Mielnik-Niedzielska, Anna Kasprzyk, Joanna Boguslawska

Summary: Adenoids, part of Waldeyer's ring, are lymphoid tissues located at the junction of the roof and posterior wall of the nasopharynx. They play a crucial role in immune system development and serve as the first line of defense against respiratory and digestive antigens. The causes of adenoid hypertrophy are not fully understood, but they are likely associated with immune reactions, infections, environmental exposures, and hormonal or genetic factors. This review aims to summarize current knowledge about adenoid hypertrophy in children and its associated diseases. Adenoid hypertrophy has various clinical manifestations in pediatric patients and is often accompanied by comorbidities.

Endosomal signaling via cAMP in parathyroid hormone (PTH) type 1 receptor biology

Karina A. Pena, Sofya Savransky, Breanna Lewis

Summary: Compartmentalization of GPCR signaling is an emerging topic that emphasizes the importance of spatial bias in signaling for physiological relevance. PTH1R was the first GPCR discovered to signal via cAMP from endosomes, challenging the conventional model of GPCR signaling. The location of cAMP generation determines the physiological outcomes of GPCR signaling.

Blocking the interaction between circTNRC18 and LIN28A promotes trophoblast epithelial-mesenchymal transformation and alleviates preeclampsia

Li-Li Chen, Ya-Qin Li, Zhi-Hui Kang, Xuan Zhang, Su-Yan Gu, Na Wang, Xue-Yan Shen

Summary: Defects in trophoblastic EMT caused by dysregulation of circTNRC18's interaction with LIN28A play a vital role in the development of preeclampsia. LIN28A overexpression suppresses circTNRC18-mediated inhibition of trophoblast migration, invasion, and EMT, while LIN28A knockdown promotes them. Furthermore, circTNRC18 regulates the intracellular distribution of LIN28A and the expression of insulin-like growth factor II, affecting cell migration and invasion. Targeting the circTNRC18-LIN28A regulatory axis may provide a novel treatment approach for preeclampsia.
