GroEL PCR-RFLP - An efficient tool to discriminate closely related pathogenic Vibrio species

Vibrio sp. are autochthonous to marine and estuarine waters. Several species of Vibrio are pathogens. It is of utmost importance to detect and discriminate the Vibrio sp. that are often involved in food and water borne infections. Since 16S rRNA based identification has limited utility in differentiating the closely related pathogenic species from non pathogenic species, we have evaluated the discriminatory power of groEL PCR-RFLP for identification of closely related Vibrio sp. Accordingly, in the current study, the efficiency of groEL PCR-RFLP for detection and accurate differentiation of known pathogens among Vibrio sp. such as V cholerae, V parahaemolyticus, V vulnificus, V mimicus, V fluvialis, V alginolyticus, V. anguillarum was evaluated. PCR amplified groEL gene fragment of each Vibrio sp. was digested separately using 5 restriction enzymes viz. Hhal, Rsal, Alul, Ddel and Mbol. The accuracy of the method was further validated by insilico restriction analysis of multiple strains of each species using NEBcutter. The method proved to be efficient for detection and differentiation of Vibrio species under study. Phylogenetic analysis also revealed groEL gene to be a better phylogenetic marker for Vibrio compared to 16S rRNA. Hence, the method can be employed for accurate detection of Vibrio sp. including the closely related species. (C) 2017 Elsevier Ltd. All rights reserved.

Abundance and diversity of diazotrophs in the surface sediments of Kongsfjorden, an Arctic fjord

Summary: This study investigated the abundance and diversity of diazotrophs in Arctic fjord sediments, revealing dominance of δ-proteobacterial diazotrophs and significant correlations with TOC/TN ratio and total organic carbon content. Glacier runoff introducing low nitrogen content organic matter enhances the abundance and diversity of nitrogen-fixing microorganisms in the fjord sediments.

Bacterial community structure and functional profiling of high Arctic fjord sediments

Summary: Kongsfjorden, an Arctic fjord, is experiencing changes in bacterial community structure of sediments due to glacier melt and Atlantification driven by accelerated warming in the Arctic. The dominant phyla in the sediments are Proteobacteria, Bacteroidetes, and Epsilonbacteraeota, with alpha- and gamma-Proteobacteria class dominating in surface sediments and delta-Proteobacteria in subsurface sediments. Total organic carbon is suggested to be a major factor affecting bacterial diversity, and bacterial communities are predicted to be involved in the degradation of complex organic compounds and biosynthesis of secondary metabolites.

Microcosm studies on the survival of Escherichia coli in the Kongsfjorden, an Arctic fjord

Summary: Microcosm experiments were conducted to study the relative survival of two Escherichia coli strains in the Kongsfjorden water during the 2017 Arctic summer. Biotic factors and solar radiation were found to be most effective in removing E. coli from the water, while the chemical composition of the fjord water supported extended survival of the bacteria.

Efficacy of a Closed Water Depuration System with Charcoal filter on the Bacteriological quality of Villorita cyprinoides var. cochinensis (Hanley, 1866)

Survival Kinetics of Vibrio Species in a Tropical Estuary along the Southwest coast of India - as a function of selected Environmental Factors
