Curcumin-mediated anti-microbial photodynamic therapy against Candida dubliniensis biofilms

The purpose of this study was to evaluate the effectiveness of anti-microbial photodynamic therapy (aPDT) mediated by curcumin (Cur) associated with LED light against biofilms of Candida dubliniensis, and further, investigate cellular uptake and drug penetration through the biofilms under confocal laser scanning microscopy (CLSM). Four C. dubliniensis strains were tested: three clinical isolates from HIV-positive patients and one reference strain (CBS 7987). Biofilms were treated with three Cur concentrations (20.0, 30.0, and 40.0 mu M). All samples were incubated in the dark for 20 min and exposed to a 5.28 J/cm(2) of LED light fluence. Additional samples of each strain were treated either with Cur or LED light only. Control samples had neither Cur nor light. After aPDT, results were read using the XTT salt reduction method. The data were statistically analyzed by two-way ANOVA followed by Games-Howell post-hoc test (alpha = 0.05). Confocal laser scanning microscopy was used to verify both the uptake of Cur by yeast cells and its penetration through the biofilm. The results showed that aPDT promoted significant reduction on the metabolism of the biofilm-organized cells of C. dubliniensis. Further, while Cur was rapidly taken up by C. dubliniensis cells, a longer time interval was required to allow Cur penetration into biofilm cells. Based on these results, aPDT associating LED and Cur presents promising potential on fungal control of biofilms of C. dubliniensis.

Impact of light-activated curcumin and curcuminoids films for catheters decontamination

Summary: Trans-resveratrol, although effective in promoting dermatological effects, has low solubility and bioavailability due to its high crystallinity. Solid dispersions were developed to address this issue, however, their powdery form makes cutaneous controlled drug delivery unfeasible. In this study, polyvinylpyrrolidone (PVP) films were utilized to develop a controlled delivery system for treating inflammation and bacterial infections associated with atopic dermatitis. The results showed that the developed system was capable of sustained release, good retention in the skin, and exhibited low cytotoxicity. Moreover, it effectively reduced inflammation and inhibited the growth of Staphylococcus aureus.

Liquid crystal precursor system as a vehicle for curcumin-mediated photodynamic inactivation of oral biofilms

Summary: This study demonstrates the antimicrobial efficacy of a curcumin-loaded liquid crystal precursor for photodynamic inactivation of dental biofilms. The results show a significant reduction in viable colonies when using the curcumin-loaded liquid crystal precursor compared to the control group. Hierarchical linear regression analysis suggests that individual characteristics may affect the efficacy of photodynamic inactivation.

Fragment reattachment or direct restoration? An in vitro study

Summary: This study aimed to evaluate the influence of different bonding materials, including pre-heated composite resin, on the fracture resistance of teeth subjected to the reattachment technique compared with a direct composite resin restoration. The results showed that the tooth reattachment technique using pre-heated composite resin showed fracture resistance values similar to those of the sound tooth group, and no difference was found between the fragment reattachment and direct composite resin techniques.

The results may lead to changes in the clinical approaches of endodontic therapy for teeth with apical periodontitis and reveal the potential of complementary approaches for enhanced root canal system decontamination during preparation.

Self-reported periodontitis association with impaired smell and taste: A multicenter survey

Summary: This study investigated the association between self-reported periodontitis and the senses of taste and smell among employees of one Danish and two American universities. Data were collected through a digital survey, including 1239 individuals from Aarhus University - Denmark, the University of Iowa, and the University of Florida - USA. Self-reported periodontitis was the exposure, and the perceived senses of taste and smell were measured using a visual analog scale (VAS). Self-perceived halitosis was the mediator. Confounders were adjusted. The total effect was decomposed into direct and indirect effects using a counterfactual approach.
