Platelet activation by SARS-CoV-2 implicates the release of active tissue factor by infected cells

Summary: Platelets are hyperactivated in COVID-19, but the mechanisms behind platelet activation by SARS-CoV-2 are not well understood. This study found that tissue factor (TF) produced by infected cells can activate platelets. Virions and extracellular vesicles (EVs) were also identified, and TF activity was associated with either the virus or the EVs. Blocking this pathway may interfere with platelet activation and the coagulation characteristics of COVID-19.

Fibrinogen, Fibrin, and Fibrin Degradation Products in COVID-19

Summary: SARS-CoV-2 infection can lead to severe pathological changes including significant activation of the clotting system and thrombus formation, resulting in abnormal fibrinogen and fibrin degradation. Fibrinogen and the fibrin degradation product D-dimer may serve as biomarkers for poor prognosis in COVID-19. Emerging studies suggest that fibrinogen plays an active role in the pathogenesis of COVID-19.

Summary: Oral contraceptive (OC) use increases the risk of venous thromboembolism. This study investigates the effect of OC hormones on endothelial cell (EC) procoagulant activity and the potential interaction between OC hormones and estrogen receptors and inflammation. The results show that the OC hormones ethinyl estradiol and drospirenone do not directly enhance the thrombin generation potential of primary ECs in vitro.

Summary: This study evaluated the satisfaction of haemophilia A patients who chose dispensation from a community pharmacy or a hospital pharmacy. The results showed that patients were more satisfied with dispensation from a community pharmacy compared to a hospital pharmacy. Accessibility to treatment was improved, travel times were significantly reduced, and patients reported higher overall satisfaction and stronger relationships with the pharmacists.

Summary: This study aimed to understand the (patho) physiological actions of fibrinogen by characterizing the genetic contribution to its synthesis. By inducing HepG2 cells to express fibrinogen and observing the effects of small interfering RNA knockdown of different candidate genes, several genes interacting with fibrinogen production were identified. This approach can reveal cellular mechanisms of fibrinogen and fibrin(ogen)-mediated (patho) physiological mechanisms.

Tissue factor pathway inhibitor is a potential modifier of bleeding risk in factor XI deficiency

Summary: This study suggests that tissue factor pathway inhibitor (TFPI) may play a role in modifying the bleeding risk in individuals with factor XI (FXI) deficiency. In vitro experiments showed that inhibition of TFPI enhanced clot formation and reduced fibrinolysis in FXI-deficient plasma, while increased TFPI had the opposite effects. Higher TFPI concentrations were observed in FXI-deficient individuals with bleeding compared to controls, and total plasma TFPI correlated with clotting and fibrinolysis parameters associated with bleeding risk. These findings indicate that TFPI may be a potential modifier of bleeding risk in FXI-deficient individuals.
