Summary: This study investigated ten samples containing promestriene (PRMS), including pure active pharmaceutical ingredient (API) and nine binary mixtures with commonly used excipients. The necessity of this study lies in the lack of reported physicochemical screening of PRMS in the literature. Additionally, the discontinuation of promestriene containing formulations in the Romanian pharmaceutical market since 2015, which caused health problems to a large number of patients, further supports the importance of this study. The results revealed that PRMS is compatible with all selected excipients, as confirmed by spectroscopy, X-ray diffraction patterns, and thermal analysis.

Summary: This study synthesized entirely S-protected thiolated hydrox-yethylcellulose (HEC) with low and high viscosity, as well as thiolated poly-L-lysine (poly-L-Lys) for dual-acting hydrogels. The thiomers showed lower cytotoxicity and significantly increased dynamic viscosity.

Summary: The compatibility of drugs with excipients is crucial for the stability of pharmaceutical formulations. Traditional analytical tools and machine learning can be used to identify drug-excipient interactions and develop predictive tools for incompatibility. In this study, a machine learning model (DE-Interact) was trained using PubChem Fingerprint as compounds' descriptors and achieved high accuracy in predicting incompatibilities.

Summary: This study investigated the physical-chemistry properties of BNZ/MGs mixtures under different processing conditions (maceration, ball milling, and melting) to guide the development of new solid formulations. The results showed that GML was more effective in improving the solubility of BNZ due to its malleable structure, less hydrophobic nature, and greater interaction with BNZ. Regardless of the processing method, the drug remained crystalline, and the presence of spherulites was observed.

Summary: This research aimed to design and characterize new hydrogels and spongious matrices for wound dressings. The compatibility of biopolymers was evaluated through rheological and spectroscopic analyses, and the stability of the matrices was studied in terms of morphology, thermal properties, and biodegradation. The results showed that the dressings had proper swelling ability, stability, and biocompatibility.

Summary: The study found that different tablet excipients have an impact on the physical form and chemical stability of levothyroxine sodium pentahydrate, including hygroscopic and acidic excipients. HPLC analysis revealed that certain excipients caused pronounced chemical decomposition of LSP, while others did not affect its stability under specific storage conditions. Excipient properties such as hygroscopicity and microenvironmental acidity were identified as critical determinants of the physical and chemical stability of LSP in a drug product.

Summary: In this study, cellulose-based core-shell particles were developed using the Layer-by-Layer (LbL) technique by alternating two cellulose derivatives onto a cationic vesicular template. The size of the particles could be tuned by controlling the number of layers deposited, with pH having a significant effect on the process. This work demonstrates the electrostatic driving force for LbL deposition and the potential advantages of using biobased containers for pharmaceutical and biomedical applications.

Summary: This study evaluated the early immune risk associated with impurities in trehalose, a pharmaceutical excipient, using multiple models. The results showed that nanoparticle impurities in trehalose can elicit an immune response in vitro, which may be related to the impurities and not the excipient itself. Assessing the immune risk of impurities in trehalose is important for the safety of high-risk administration routes.

Summary: N-nitrosamines are potential carcinogens, and there has been a regulatory framework established for risk assessment, testing, and mitigation of these contaminants in drug products. One approach to prevent the formation of N-nitrosamines during drug manufacturing and storage is to incorporate nitrite scavengers into the formulation. Various molecules, including antioxidant vitamins, amino acids, and other antioxidants used in food or drugs, have been tested for their potential to mitigate N-nitrosamine formation in drug products. This review article provides a comprehensive overview of key considerations for incorporating nitrite scavengers in oral drug formulations.

Summary: The crystal structures and thermal characteristics of two polymorphs of methylethanolammonium 2-chloro-4-nitrobenzoate were analyzed using experimental and computational methods. The study focused on the synthesis of the polymorphs, structural characterization, intermolecular interactions, thermodynamic stability, and thermal decomposition kinetics. The results revealed distinct crystal packing arrangements and different numbers of independent molecules pairs in the asymmetric crystal unit. The alpha polymorph had a more stable lattice than the beta polymorph at high temperatures. The thermodynamic stability was determined by measuring melting points and heats of fusion. The alpha polymorph also exhibited higher activation energies compared to the beta polymorph in the thermal decomposition processes.

Summary: The objectives of this study were to formulate and evaluate medicated oral jellies containing Ambroxol HCl, the most commonly used mucolytic agent for chronic bronchitis, as well as medicated oral jellies containing Ibuprofen, a recognized nonsteroidal anti-inflammatory drug. The medicated jellies included different gelling agents such as gelatin, sodium alginate, and pectin. Compatibility studies were conducted to assess the interaction between Ibuprofen or Ambroxol and each component of the medicated jellies. Analysis was performed using FTIR-UATR and thermogravimetry.

Summary: The stability of APIs and pharmaceutical formulations is crucial in pharmaceutical research and technology. Various factors such as light, air exposure, temperature, humidity, formulation composition, and packaging materials can affect the quality and stability of APIs and finished pharmaceutical products. This study analyzed the degradation kinetics of the antihypertensive drug moxonidine in different solid mixtures using FTIR spectroscopy and thermoanalytical methods. The results showed that excipients have a stabilizing effect on moxonidine, but the decomposition mechanism is complex.

Summary: The current nanomedicinal approach combines medicinal plants and nanotechnology to create new scaffolds with enhanced bioavailability, biodistribution, and controlled release. This study presents an innovative method for encapsulating herb in nanosized chitosan matrices, and investigates the properties and therapeutic effects of these nanocarriers. The results suggest that this new antioxidant vehicle has potential as a promising candidate in tumor therapeutic strategy.
