Synthesis of All Possible Canonical (3′-5′-Linked) Cyclic Dinucleotides and Evaluation of Riboswitch Interactions and Immune-Stimulatory Effects

The cyclic dinucleotides (CDNs) c-di-GMP, c-di-AMP, and c-AMP-GMP are widely utilized as second messengers in bacteria, where they signal lifestyle changes such as motility and biofilm formation, cell wall and membrane homeostasis, virulence, and exo-electrogenesis. For all known bacterial CDNs, specific riboswitches have been identified that alter gene expression in response to the second messengers. In addition, bacterial CDNs trigger potent immune responses, making them attractive as adjuvants in immune therapies. Besides the three naturally occurring CDNs, seven further CDNs containing canonical 3'-5'-linkages are possible by combining the four natural ribonucleotides. Herein, we have synthesized all ten possible combinations of 3'-5'-linked CDNs. The binding affinity of novel CDNs and GEMM riboswitch variants was assessed utilizing a spinach aptamer fluorescence assay and in-line probing assays. The immune-stimulatory effect of CDNs was evaluated by induction of type I interferons (IFNs), and a novel CDN c-AMP-CMP was identified as a new immune-stimulatory agent.

Summary: In recent years, the adverse effects of sunscreens on the environment and biology have become a major concern. To improve sunscreen safety, researchers have developed a bionic photoprotective material, CS-SA-PDA nanosheet, by simulating the photoprotective mechanisms of skin and plant leaves. The nanosheet effectively shields against UVB and UVA, improves photostability, reduces skin permeability, and can scavenge photoinduced free radicals. It also has no skin irritation and is excellent against skin photodamage, making it an ideal skin photoprotective material.

Rational design of dual-functional surfaces on polypropylene with antifouling and antibacterial performances via a micropatterning strategy

Summary: In this study, a dual-function micropatterned polypropylene (PP) surface with antifouling and antibacterial properties was successfully constructed through plasma activation, photomask technology, and ultraviolet light-induced graft polymerization. Two different micropatterning structures were prepared using antifouling and antibacterial agents, and the performance of the resulting materials was thoroughly characterized. The results showed that the dual-functional PP materials exhibited good resistance against protein and platelet adhesion, as well as high antibacterial activity.

An ATP-Cu(ii) catalyst efficiently catalyzes enantioselective Michael reactions in water

Summary: In this study, a single nucleotide-based catalyst (ATP·Cu2+) was successfully constructed and demonstrated to catalyze enantioselective Michael reactions in aqueous media with high reactivities and good enantioselectivities up to 83% ee. The enantioselective catalytic performances of ATP·Cu2+ originate from specific binding between ATP and Cu2+ ions that exhibit both pH-dependent and temperature-dependent behaviors. The ATP·Cu2+ catalyst has been successfully applied to a wide spectrum of enone substrates and carbon nucleophiles.
