Introduction: The association between ischemic stroke and genetic polymorphisms of methylenetetrahydrofolate reductase (MTHFR; 677C>T and 1298A>C), endothelial nitric oxide synthase (eNOS; -786T>C, +894G>T, and variable number tandem repeat [VNTR]), phosphodiesterase 4D (PDE4D; SNPs 83 and 87), angiotensin-converting enzyme (ACE) I/D, angiotensinogen (AGT) 235M>T, paraoxonase 1 (PON1) 192Q>R, and apolipoprotein E (ApoE) epsilon 2 epsilon 3 epsilon 4 remains inconclusive. Therefore, this updated meta-analysis aimed to clarify the presumed influence of genetic polymorphisms on ischemic stroke by meta-analyzing the comprehensive coverage of all individual association studies. Methods: All case-control studies published in different languages such as English, Japanese, Korean, Spanish, Chinese, Hungarian, Ukrainian, or Russian were identified from databases. The pooled odds ratios (ORs) and 95% confidence intervals (CIs) were calculated via fixed-and random-effect models. Sensitivity analysis, heterogeneity test, Hardy Weinberg Equilibrium, and Egger's regression analyses were performed in this study. Results: A total of 490 case-control studies with 138,592 cases and 159,314 controls were included in this meta-analysis. Pooled ORs from all the genetic models indicated that MTHFR 677TT and 1298CC, eNOS +894TT and VNTR, PDE4D SNP 83, ACE DD, AGT 235TT, PON1 192RR, and ApoE epsilon 4 polymorphisms were increasing the risks of ischemic stroke. Nevertheless, PDE4D SNP 87 and eNOS -786T>C polymorphisms are not associated with ischemic stroke risks. Conclusions: Hence, the evidence from this meta-analysis concluded that MTHFR (677C>T and 1298A>C), eNOS (+894G>T and VNTR), PDE4D SNP 83, ACE I/D, AGT 235M>T, PON1 192Q>R, and ApoE epsilon 2 epsilon 3 epsilon 4 polymorphisms predispose individuals to ischemic stroke.

Underestimation of Heritability across the Molecular Layers of the Gene Expression Process

Jihye Ryu, Chaeyoung Lee

Summary: The study revealed significant heritability underestimation for molecules from an infinitesimal model in mixed model analysis, emphasizing the importance of using an optimal number of nucleotide variants to accurately estimate heritability and identify expression quantitative trait loci (eQTLs). Additionally, a heterogeneous covariance structure based on gene identity and the molecular layers of the gene expression process is essential for better understanding polygenic effects and minimizing errors in identifying eQTLs.

Identification and Interpretation of eQTL and eGenes for Hodgkin Lymphoma Susceptibility

Yeeun An, Chaeyoung Lee

Summary: In this study, transcriptome-wide analysis of expression quantitative trait loci (eQTL) was conducted to identify target genes associated with Hodgkin lymphoma (HL) genome-wide association studies (GWAS) signals. A total of 80 eGenes were identified to be associated with 20 HL GWAS signals. Enrichment analysis revealed the functions of these eGenes involved apoptosis, immune responses, and cytoskeletal processes. Further investigation on two specific eGenes suggested that their minor alleles could be genetic risk factors for HL susceptibility.

Adam Vacek, Grant Mair, Philip White, Philip M. Bath, Keith W. Muir, Rustam Al-Shahi Salman, Chloe Martin, David Dye, Francesca M. Chappell, Ruediger von Kummer, Malcolm Macleod, Nikola Sprigg, Joanna M. Wardlaw

Summary: This study evaluated the performance of e-ASPECTS software in delineating the extent of intracranial hemorrhage (ICH) on brain CT scans. Summary: This study aimed to evaluate the efficacy and safety of endovascular treatment (EVT) in primary and EVT-induced secondary medium-vessel occlusions (MeVO). The results showed that primary MeVO had poorer outcomes in terms of complications, EVT efficacy, and the proportion of patients with a modified Rankin Scale (mRS) of 0-2 at 3 months compared to exclusive large-vessel occlusion (LVO). However, EVT-induced secondary MeVO showed similar safety and efficacy outcomes as exclusive LVO and had better outcomes in terms of mRS 0-2 compared to primary MeVO.

Summary: This study assessed the geographic accessibility of stroke rehabilitation facilities in Tennessee. The study found that rural counties had significantly longer travel distances and times compared to urban counties. Counties with high and medium stroke prevalence also had higher travel times and distances compared to low stroke prevalence counties. Therefore, addressing transportation barriers and implementing tele-rehabilitation are crucial for improving access to stroke aftercare in these areas.

Summary: The study aimed to investigate the relationship between serum alkaline phosphatase (ALP) levels and the severity and prognosis of intracerebral hemorrhage (ICH). The results showed that serum ALP levels in ICH patients were significantly elevated and correlated with hematoma volume and Glasgow coma scale (GCS) score. High serum ALP levels may serve as a potential predictor for the severity and unfavorable prognosis of ICH.
