Rapid Characterization of the Human Breast Milk Lipidome Using a Solid-Phase Microextraction and Liquid Chromatography Mass Spectrometry-Based Approach

Human breast milk (HBM) is a biofluid consisting of various biomolecules such as proteins, lipids, carbohydrates, minerals and bioactive substances. Due to its unique and complex composition, HBM provides not only nutritional components required for the growth of the infant, but also additional protection against infections. Global insight into the composition of HBM is crucial to understanding the health benefits infants receive from breastfeeding and could be used to improve the composition of milk formula for babies that cannot be breastfed. To improve global profiling of the HBM lipidome, a new analytical approach based on solid phase microextraction (SPME) and liquid chromatography mass spectrometry (LC-MS) was developed. The new extraction method allows for the rapid and simple extraction of a broad range of lipids directly from HBM samples. Moreover, the optimized two-step lipid extraction protocol ensures high lipidome coverage without using toxic solvents such as chloroform. The use of liquid chromatography quadrupole time-of-flight mass spectrometry (LC-QTOF-MS) and an automated search of a lipid database allows comprehensive identification of the lipids contained in HBM. The demonstrated analytical approach based on SPME sample preparation and LC-QTOF-MS is rapid, free of toxic solvents, and suited for the qualitative analysis of the HBM lipid composition.

Summary: This study focused on the genotoxicity of pharmaceuticals from different therapeutic groups, both in mixtures and individual solutions, under different environmental conditions. The results showed that the addition of inorganic ions mainly reduced genotoxicity, while changes in the pH of pharmaceutical solutions significantly affected genotoxicity. Synergistic interactions were more commonly observed among mixtures.

Comparative evaluation of different methods for determining phytochemicals and antioxidant activity in products containing betalains-Verification of beetroot samples

The Influence of Storage on Human Milk Lipidome Stability for Lipidomic Studies

Summary: The study found that storage of human milk samples at 4 degrees Celsius for 4 days and subjecting them to three freeze-thaw cycles can lead to changes in lipid content. Differences in levels of certain lipid species between samples stored at -20 degrees Celsius and -80 degrees Celsius were not statistically significant, and individual variance remained consistent regardless of sample storage conditions. Storing HM samples at -20 degrees Celsius for up to 3 weeks and at -80 degrees Celsius for up to 12 weeks ensures sample lipidome stability.

A comprehensive lipidomic analysis of oilseeds using LC-Q-TOF-MS and dispersive micro-solid phase (D-?-SPE) extraction techniques

Summary: Growing public health awareness has led to increased demand for natural products with positive health benefits. This study conducted a comprehensive analysis and comparison of the lipid profiles of seven oilseeds using D-mu-SPE and LC-Q-TOF-MS. The results showed that flax, hemp, and chia seeds had similar lipid content, while black cumin, black and white sesame seeds had a similar lipid composition with more saturated fatty acid substituents. ALA and LA were present in all lipid classes.
