Summary: In this study, a functionalized vinyl-linked 2D fluorescent covalent organic framework (COF) (TMT-TA-COOH) with good water dispersibility and enhanced binding ability was constructed by post-synthetic modification (PSM) of a TMT-TA COF. The TMT-TA-COOH demonstrated high sensitivity in the quantitative identification of 5-hydroxytryptamine (5-HT) and 5-hydroxyindole-3-acetic acid (5-HIAA) in water, as well as specific recognition of 5-HT in serum and 5-HIAA in urine. Additionally, TMT-TA-COOH showed promise as a tool for early detection of neurological diseases and biomarkers for carcinoid tumors, as well as latent fingerprint (LFP) identification.

Summary: The hormone cholecystokinin (CCK) is released by duodenal enteroendocrine cells after a meal and circulates in the low picomolar range. Current methods for detecting CCK suffer from sensitivity issues and cross-reactivity problems. Therefore, a new liquid chromatography and mass spectrometry-based method was developed to measure CCK-derived peptides in cell culture supernatants. This method proved effective in detecting the peptide ppCCK(21-44) in human plasma and secretion buffer solutions.

Summary: Based on coordination post-synthetic modification, a Tb3+ functionalized ME-PA (Tb@1) is prepared, which can emit brilliant green fluorescence through ligand-to-mental charge transfer-assisted energy transfer (LMCT-ET) process from ME-PA to Tb3+ ions. Tb@1 can simultaneously distinguish Tryptophan (Try) and its metabolite 5-hydroxyindole-3-acetic acid (5-HIAA), two effective indicators for depression, in ratio and colorimetric mode. This work offers a novel approach to modulate logic circuits based on ML-assisted HOF fluorescent sensor, with promising application for a precise and pictorial depression diagnosis.

Summary: This study investigates the possible mechanism involved in the development of ethanol withdrawal syndrome (EWS) by administering a 5-hydroxytryptamine (5-HT) 1b/1d receptor agonist, Zolmitriptan. The results reveal that Zolmitriptan can reverse the behavioral changes induced by EWS and reduce anxiety levels in EWS rats. Furthermore, Zolmitriptan stimulates the level of gamma-aminobutyric acid (GABA) and improves the levels of other neurochemicals in the brain of EWS rats.

Summary: In this study, the therapeutic effect of MJGT_EE on IBS-C was investigated in a rat model. The results showed that MJGT_EE increased FWC and CRD volume, promoted gastric emptying and small intestinal propulsion. It also regulated the expression of proteins related to the 5-HT pathway, enhanced the diversity of gut microbiota, and increased the proportion of beneficial bacteria.

Summary: The use of trapped ion mobility spectrometry (TIMS) in conjunction with liquid chromatography (LC) and electrospray ionization tandem mass spectrometry (MS/MS) was explored for the analysis of PDE-5 inhibitor analogs. Six out of eight compounds were resolved by LC and TIMS, while the remaining two isomers were distinguished by unique product ions in the MS/MS spectrum. The LC separation corresponded to differences in substitution on the piperazine moiety of the PDE-5 inhibitors, while the TIMS separation corresponded to the position of a nitrogen atom in the fused ring region of the molecules.

Summary: This study assessed the application of 5-hydroxyindole acetic acid as a biochemical marker of neuroendocrine tumours in plasma and serum. The results showed that plasma and serum 5-hydroxyindole acetic acid had similar sensitivity and specificity compared to the urinary assay. Therefore, plasma and serum samples can be interchangeably used for this testing.

Summary: In this study, the quality of Yiqing tablet was comprehensively evaluated using multi-component quantification, multi-dimensional fingerprint construction, and antioxidant activity analysis. The results showed that the sample had 25 components with antioxidant activity according to the HPLC fingerprint and most bands of FTIR also showed antioxidant activity. Based on the evaluation results of HPLC and FTIR fingerprint, all samples were classified as first level, demonstrating the consistency of sample quality. This study provides a new idea for achieving comprehensive quality evaluation of traditional Chinese medicines.

Summary: This work presents a simple, cost-effective, and environmentally friendly method for determining the content of multiple elements in medicinal herbs using ICP OES. The optimized experimental parameters result in low dissolved organic carbon content and residual acidity concentration. The accuracy of the method is confirmed through the analysis of certified reference materials, and its environmental friendliness is validated using the AGREE metric. Principal component analysis is used to classify the medicinal herbs based on their inorganic constituent concentrations.

Summary: This study investigated the cellular uptake of low, medium and high molecular weight polyethylene glycol (PEG) in MCF-7 cells using UPLC-MS/MS and CID techniques. The results showed that PEG600, PEG2K, PEG4K and PEG10K exhibited low toxicity and had limited cellular entry at concentrations of 5-1200 μg/mL. Additionally, the environmental friendliness of the developed analytical methods was evaluated.

Summary: This study employed network pharmacology-based analysis and reverse molecular docking to investigate the molecular targets and pathways associated with gynecological disorders and the potential therapeutic effects of fenugreek constituents. The results highlighted the potential of steroidal saponins, isoflavones, and flavones as active constituents of fenugreek with implications for lactation, reproductive processes, and estrogenic activity. Animal model study further supported the presence of these bioactive compounds and their effects on body weight, serum prolactin levels, serum estradiol levels, and ovarian histological structure.
