A systematic meta-analysis of the association of Neuregulin 1 (NRG1), D-amino acid oxidase (DAO), and DAO activator (DAOA)/G72 polymorphisms with schizophrenia

The glutamate hypothesis of schizophrenia is related to the proposed dysregulation of d-amino acid oxidase (DAO), DAO activator (DAOA)/G72, and Neuregulin 1 (NRG1) genes. Genetic studies have shown significant associations between DAO, DAOA, NRG1 single-nucleotide polymorphisms (SNPs), and schizophrenia. The systematic literature search yielded 6, 5, and 18 new studies for DAO, DAOA, and NRG1 published after 2011 and not included in the previous SchizophreniaGene (SZGene) meta-analysis. We conducted meta-analyses of 20, 23, and 48 case-control studies, respectively, to comprehensively evaluate the association of 8 DAO, 12 DAOA, and 14 NRG1 SNPs with schizophrenia. The updated meta-analyses resulted in the following findings: the C-allele of DAO rs4623951 was associated with schizophrenia across all pooled studies [Odds ratio (OR) = 0.88, 95% confidence interval (CI) = 0.79-0.98, p = 0.02, N = 3143]; however, no new reports could be included. The G-allele of DAOA rs778293 was associated with schizophrenia in Asian patients (OR = 1.17, 95% CI = 1.08-1.27, p = 0.00008, N = 6117), and the T-allele of DAOA rs3916971 was associated with schizophrenia across all studies (OR = 0.84, 95% CI = 0.73-0.96, p = 0.01, N = 1765). Again, for both SNPs, no new eligible studies were available. After adding new reports, the T-allele of NRG1 SNP8NRG241930 (rs62510682) across all studies (OR = 0.95, 95% CI = 0.91-0.997, p = 0.04, N = 22,898) and in Caucasian samples (OR = 0.95, 95% CI = 0.90-0.99, p = 0.03, N = 16,014), and the C-allele of NRG1 rs10503929 across all studies (OR = 0.89, 95% CI = 0.81-0.97, p = 0.01, N = 6844) and in Caucasian samples (OR = 0.89, 95% CI = 0.81-0.98, p = 0.01, N = 6414) were protective against schizophrenia. Our systematic meta-analysis is the most updated one for the association of DAO, DAOA, and NRG1 SNPs with schizophrenia.

Summary: This study aimed to reveal the neurobiological characteristics of disruptive behavior disorders (DBD) by integrating individual deviation patterns from multiple imaging modalities. The results showed that DBD patients exhibited increased age-related deviations in the amygdala, suggesting a possible maturational delay. Furthermore, the study identified neural signatures associated with aggression, including the default mode network (DMN), striatum, and amygdala.

Summary: In order to identify potential novel medications for child and adolescent mental health problems, we conducted a systematic search on clinical trial databases from 2010 to 2022. We included phase 2 or 3 randomized controlled trials (RCTs) of medications without regulatory approval in the US, Europe or Asia, and also RCTs of dietary interventions/probiotics. We found 234 ongoing or completed RCTs, with varying results on primary outcomes.

Summary: This study compares the cognitive and affective empathy abilities of youth with conduct disorder (CD) and autism spectrum disorder (ASD), and investigates the influence of comorbid Callous-Unemotional (CU) traits on empathy. Results show that the ASD group has lower parent-reported empathy scores and self-reported cognitive empathy scores, while the CD group has lower parent-reported cognitive empathy scores. However, when accounting for the influence of CU traits, deficits in affective empathy for ASD and deficits in cognitive empathy for CD were no longer found.

Summary: The study found that Mind My Mind (MMM) cognitive behavioral therapy is effective in treating common emotional and behavioral mental health problems in youth, but not all individuals respond satisfactorily to treatment. The analysis revealed that youths with any mental disorder at baseline, comorbidity, and longer duration of untreated mental health problems showed superior treatment benefits. This highlights the importance of community-based programs like MMM for youth with substantial mental health problems.

Associations of n-3 polyunsaturated fatty acid status and intake with paediatric major depressive disorder in Swiss adolescents: A case-control study

Summary: This study found an association between n-3 polyunsaturated fatty acid (PUFA) status and intake and paediatric major depressive disorder (pMDD) in Swiss adolescents. The composition of red blood cell (RBC) fatty acids showed lower levels of EPA and DHA in pMDD cases, as well as a lower n-3 PUFA index and higher n-6/n-3 PUFA ratio, which were associated with increased risk for pMDD.

Antipsychotic effects on anthropometric outcomes in anorexia nervosa: a retrospective chart review of hospitalized children and adolescents

Summary: This study found that the use of antipsychotic medication in the treatment of anorexia nervosa in youth did not significantly improve weight restoration compared to those not treated with antipsychotics. Comparison of clinical outcomes in patients with schizophrenia following different long-acting injectable event-driven initiation strategies

Summary: This retrospective study evaluated the benefit of different long-acting injectable (LAI) initiation strategies based on timing of behavioral and clinical events among Medicaid beneficiaries with schizophrenia. The study found that initiating LAIs prior to nonadherence to oral antipsychotics (OAPs) or occurrence of schizophrenia-related inpatient/ER visits was associated with fewer all-cause inpatient days and ER visits. Therefore, the LAI initiation strategy plays an important role in improving clinical outcomes for patients with schizophrenia.

Pharmacological interventions for antipsychotic-related sialorrhea: a systematic review and network meta-analysis of randomized trials

Summary: This study conducted a network meta-analysis on antipsychotic-induced sialorrhea. The results showed that dopamine receptor antagonists, metoclopramide, and sulpiride were effective in treating sialorrhea, while antihistamines and atropine showed no significant effect on nocturnal sialorrhea. This study provides some guidance for the treatment of antipsychotic-related sialorrhea.
