Preussilides A-F, Bicyclic Polyketides from the Endophytic Fungus Preussia similis with Anti proliferative Activity

Six novel bioactive bicyclic polyketides (1-6) were isolated from cultures of an endophytic fungus of the medicinal plant Globularia alypum collected in Batna, Algeria. The producer organism was identified as Preussia similis using morphological and molecular phylogenetic methods. The structures of metabolites 1-6, for which the trivial names preussilides A-F are proposed, were elucidated using a combination of spectral methods, including extensive 2D NMR spectroscopy, high-. resolution mass spectrometry, and CD spectroscopy: Preussilides were tested for antimicrobial and antiproliferative effects, and, in particular, compounds I and 3 showed selective activities against eukaryotes. Subsequent studies on the influence of 1 and 3 on the morphology of human osteosarcoma cells (U2OS) suggest that these two polyketides might target an enzyme involved in coordination of the cell division cycle. Hence, they might, for instance, affect timing or spindle assembly mechanisms, leading to defects in chromosome segregation and/or spindle geometry.

Multiformin-Type Azaphilones Prevent SARS-CoV-2 Binding to ACE2 Receptor

Summary: Protein microarray screenings have identified fungal natural products from the azaphilone family as powerful inhibitors of SARS-CoV-2 spike protein binding to ACE2 receptors. Among them, Cohaerin F exhibited the highest potency in reducing ACE2 and SARS-CoV-2 spike protein binding by more than 50%. Further analysis led to the discovery of six new multiformins and the revision of the stereochemistry of multiformins. Cell-based infection assays revealed that Cohaerin C and Cohaerin F significantly reduced SARS-CoV-2 pseudotyped lentivirus particles infection, while Multiformin C and G almost completely abolished the infection. These findings highlight the preventive potential of multiformin-type azaphilones against SARS-CoV-2 binding to ACE2 receptors.

Studies on the Genus Pyrenopolyporus (Hypoxylaceae) in Thailand Using a Polyphasic Taxonomic Approach

Summary: In this study, hypoxylaceous specimens collected from several sites in Thailand over the past two decades were examined to determine their affinity to the genus Pyrenopolyporus. The researchers used a combination of morphological, metabolomic, and molecular techniques to describe and illustrate five novel species and a new record for the country. Proteomic profiles of the fungi were also provided for the first time, using matrix associated laser desorption ionization-time-of-flight mass spectrometry (MALDI-TOF/MS).

Secondary metabolites of Diaporthe cameroonensis, isolated from the Cameroonian medicinal plant Trema guineensis

Summary: A new species of Diaporthe cameroonensis was isolated from the fresh root of Trema guineensis (Ulmaceae). The strain was fermented and extracted, resulting in the isolation of a hemiketal polyketide and an acetylated alternariol, as well as fifteen known secondary metabolites.

Bhushaniella gen. nov. (Cordycipitaceae) on spider eggs sac: a new genus from Thailand and its bioactive secondary metabolites

Summary: Fungal specimens parasitic on spider egg sacs were identified based on molecular phylogenetic analyses and morphological data, and a novel monotypic genus named Bhushaniella rubra was described. The fungus produces superficial perithecia and its anamorph has verticillate phialides with a slightly curved neck. The evaluation of its secondary metabolites revealed the presence of penicolinate type alkaloids, with no significant antibiotic activity or cytotoxic effects.

Cytochalasans and Their Impact on Actin Filament Remodeling

Summary: The eukaryotic actin cytoskeleton is a dynamic network composed of monomeric and filamentous forms of actin, as well as actin-binding proteins (ABPs). Small molecular inhibitors have been used to study the importance of actin filaments for cellular processes, by interfering with the interactions and turnover of the actin cytoskeleton. This review aims to summarize the available information on the structural features of cytochalasans and their effects on cell morphology and actin cytoskeleton organization.

Diaporphasines E and F: New Polyketides from the Saprotrophic Fungus Lachnum sp. IW157 Growing on the Reed Grass Phragmites communis

Summary: In this study, two polyketide metabolites were identified from the mycelial extract of a saprotrophic fungus growing on common reed grass. One of the compounds exhibited potent cytotoxicity against tested cell lines, while the other showed moderate inhibitory effects on the formation of S. aureus biofilms.

Unprecedented Antimicrobial and Cytotoxic Polyketides from Cultures of Diaporthe africana sp. nov.

Summary: Four unprecedented polyketides were isolated from the newly described endophytic fungus Diaporthe africana. The fungus was taxonomically identified using a polyphasic approach. Inhibitory Effects of the Fungal Pigment Rubiginosin C on Hyphal and Biofilm Formation in Candida albicans and Candida auris

Summary: The fungal human pathogens, Candida auris and Candida albicans, have various virulence mechanisms, including the formation of biofilms and the development of (pseudo)hyphae to protect against harsh conditions and facilitate invasion of host tissues. Increasing rates of antifungal resistance in these pathogens pose a challenge for effective treatment. This study found that rubiginosin C, derived from the ascomycete Hypoxylon rubiginosum, effectively inhibited biofilm formation and hyphal development in both C. auris and C. albicans without harming host cells. Rubiginosin C is a promising candidate for therapeutic intervention in the future.

Recent progress in the evaluation of secondary metabolites from Basidiomycota

Summary: This review summarizes recent reports on biologically active secondary metabolites from Basidiomycota in the past five years. These organisms are still largely unexplored, with a predominance of terpenoids and meroterpenoids, but also commonly encountered polyketides, alkaloids, and other nitrogen-containing metabolites. A significant number of novel compounds have been reported from African and Asian species, while there have been very few new records from American Basidiomycota and only a handful from European mushrooms. In total, over 350 new metabolites have been recorded.

Biologically active drimane derivatives isolated from submerged cultures of the wood-inhabiting basidiomycete Dentipellis fragilis

Summary: Four new drimane sesquiterpenoids were discovered in the submerged cultures of the basidiomycete Dentipellis fragilis. The compounds were characterized using advanced spectroscopic techniques and their biological activities were evaluated. One compound showed weak antimicrobial activity, while two others exhibited neurotrophic effects. This study provides new insights into the bioactive compounds produced by this group of fungi.

Lasiodipline G and other diketopiperazine metabolites produced by Lasiodiplodia chiangraiensis

Summary: In this study, two new metabolites (1 and 2) and three known compounds were isolated from the submerged cultures of L. chiangraiensis. The chemical structures of the isolated compounds were determined using NMR spectroscopic analyses and HRESIMS, and the absolute configurations of the new compounds were established using TDDFT-ECD spectra. Compound 1 showed significant cytotoxic activities against various cell lines and moderate antibacterial effects.

Siderophore conjugation with cleavable linkers boosts the potency of RNA polymerase inhibitors against multidrug-resistant E. coli

Summary: The study aimed to enhance the potency of existing antibiotics against antibiotic-resistant Gram-negative bacteria. By utilizing the microbial iron transport machinery, covalent modifications were made to improve drug translocation across the cell membrane. Cleavable linkers were designed to release the antibiotic inside the bacteria and unperturbed target binding. The conjugates showed increased antibiotic activity against multidrug-resistant E. coli compared to free rifamycin, suggesting the potential of this approach to boost the effectiveness of existing antibiotics.

Bioactive Compounds from an Endophytic Pezicula sp. Showing Antagonistic Effects against the Ash Dieback Pathogen

Summary: A fungal endophyte from the Canary Islands has been identified as an effective antagonist against the fungal phytopathogen that causes ash dieback disease. The endophyte produces various secondary metabolites, including a new compound called peziculastatin. These compounds show significant inhibitory or dispersive effects on fungal and bacterial biofilms.

Sporothioethers: deactivated alkyl citrates from the fungus Hypomontagnella monticulosa

Summary: Submerged cultivation results in the formation of new sulfur-containing sporothriolide derivatives, which show attenuated antimicrobial activity due to the presence of 2-hydroxy-3-mercaptopropanoic acid. However, these derivatives still exhibit moderate effects on the biofilms of Candida albicans and Staphylococcus aureus at lower concentrations.
