Chronic adrenergic stimulation induces brown adipose tissue differentiation in visceral adipose tissue

BackgroundRecruitment of brown adipose tissue is a promising strategy to treat obesity and Type 2 diabetes, but the physiological effects of a large amount of metabolically active brown adipose tissue in humans are unknown. Case reportIn the present paper, we report a case of massive brown adipose tissue infiltration of the visceral adipose tissue depot in a person with Type 2 diabetes with a catecholamine-secreting paraganglioma. The patient was evaluated with [18F]-fludeoxyglucose positron emission tomography/computed tomography on three occasions: pre-therapy, during -blockade and postoperatively. During surgery, biopsies of visceral and subcutaneous adipose tissue were obtained and evaluated for brown adipose tissue. At diagnosis, brown adipose tissue glucose uptake, assessed by [18F]-fludeoxyglucose-positron emission tomography, was massively increased. [18F]-fludeoxyglucose uptake was confined to known locations for brown adipose tissue, with additional uptake in the visceral adipose tissue. As a result of increased thermogenesis, resting energy expenditure was doubled. After surgical removal of the tumour, antidiabetic medicine was no longer needed, despite an 8.2-kg weight gain. ConclusionThese results show that human visceral adipose tissue holds an unprecedented potential for brown adipogenic differentiation; however, a detrimental effect on glucose metabolism persisted despite massive brown adipose tissue activity, with a doubling of resting energy expenditure.

A Persistent Left-Sided Superior Vena Cava Detected on a O-15-H2O PET/CT of the Heart

Summary: A persistent left-sided superior vena cava (PLSVC) is a rare condition with a prevalence of 0.5% in the general population. It occurs when the left anterior cardinal vein fails to regress as the ligament of Marshall during embryologic development. Although usually asymptomatic and discovered incidentally, its recognition is important due to potential complications during invasive cardiovascular procedures. In this case report, we present an incidental finding of PLSVC in a patient referred for myocardial perfusion imaging using O-15-H2O PET/CT.

Inclusion of Metabolic Tumor Volume in Prognostic Models of Bone and Soft Tissue Sarcoma Increases the Prognostic Value

Summary: This study aimed to evaluate the prognostic accuracy of combining PET parameters and circulating biomarkers in sarcoma patients. The results showed that adding PET parameters to existing prognostic models improved the prognostic value in all models. A new prognostic model, SBSpib, which includes albumin, lymphocytes, and metabolic tumor volume (MTV) as PET parameter, was developed. The study suggests that combining PET parameters and circulating biomarkers can enhance prognostic evaluation, but further validation is needed for SBSpib.

Summary: This study investigated the ability of the ESC-PTP, RF-CL, and CACS-CL models to diagnose myocardial perfusion defects (MPDs). The results showed that compared with the ESC-PTP, the RF-CL and CACS-CL models can better classify patients into a low-risk group with a low prevalence of MPDs.

Multiparametric dynamic whole-body PSMA PET/CT using [68Ga]Ga-PSMA-11 and [18F]PSMA-1007

Summary: In this study, dynamic whole-body PET imaging was used to compare the pharmacokinetics of [Ga-68]Ga-PSMA-11 and [F-18]PSMA-1007 in different tissues and lesions. The study found that the two PSMA tracers exhibited different kinetics in organs and lesions, with [F-18]PSMA-1007 showing irreversible kinetics. Multiparametric PSMA images increased target-to-background ratios but did not lead to the detection of more lesions. For quantitative whole-body Ki imaging, [F-18]PSMA-1007 should be preferred over [Ga-68]Ga-PSMA-11.

Intravenous and oral whole body ketone dosimetry, biodistribution, metabolite correction and kinetics studied by (R)-[1-11C] β-hydroxybutyrate ([11C]OHB) PET in healthy humans

Summary: This study used positron emission tomography (PET) to investigate the uptake and tissue kinetics of the ketone tracer (R)-[1-C-11]beta-hydroxybutyrate -([C-11]OHB) in the human body. The results showed that intravenous administration of [C-11]OHB resulted in high uptake in the heart, liver, and kidneys, while oral ingestion led to rapid appearance in the blood and uptake in the heart, liver and kidneys. The study suggests that [C-11]OHB may serve as a safe and non-invasive imaging tool for exploring ketone metabolism in organs and tissues.

The results of this study will be disseminated through international peer-reviewed journals.

Single time point quantitation of cerebral glucose metabolism by FDG-PET without arterial sampling

Summary: The study found strong relationships among different parameters of the input function derived from images, which enabled estimation of missing data and accurate calculation of FDG-Ki in the brain. The study also identified age-dependent aspects in the input function data, which was not previously reported in FDG pharmacokinetics. These findings support the quantitation of FDG-Ki in the brain using simplified methods with minimal error.
