Summary: BCR-ABL-independent TKI resistance in CML is mediated by FAM167A, which activates the noncanonical NF-kappa B pathway. FAM167A may serve as an important target and biomarker for BCR-ABL-independent TKI resistance.

Summary: The paper presents a morphology-based deep learning system to identify senescent cells and evaluate the effects of anti-senescent reagents. The system uses convolutional neural networks (CNN) to classify cellular senescence and successfully identifies anti-senescent drugs through morphology-based drug screening.

Summary: Safe and effective nonsteroidal anti-inflammatory drugs are necessary, and adding amino acids to cultures of microorganisms may increase the likelihood of discovering new secondary metabolites. Two new compounds, myceliostatins A and B, with the myceliothermophin structure were isolated from a methionine-enriched culture of Myceliophthora thermophila. Both compounds exhibited inhibition of LPS-induced NO production and had anti-inflammatory and antioxidant effects by suppressing NF-KB activity and reactive oxygen species production.

Summary: The study suggests that Crizotinib holds potential as a treatment for GBM, although its effectiveness is limited due to poor penetration of the blood brain barrier. Additionally, conjugation of Crizotinib with a dye showed improved inhibition of glioma cell proliferation and survival. Furthermore, IR-Crizotinib demonstrated effective tumor localization and growth inhibition in mice.

Summary: In this study, a novel targeted anticancer agent called MBP-11901 has been presented as a potential therapeutic strategy for advanced or unresectable liver cancer. The drug showed effectiveness against various human HCC cell lines and induced anticancer effects by targeting the signaling mechanisms of FLT3, VEGFR2, c-KIT, and PDGFR beta.

Summary: The study investigated the effects of three clinically established TKIs (imatinib, sunitinib, nilotinib) on KATP channel activity, revealing that these drugs can decrease KATP channel activity by interacting with the ATP-binding site on the channel. This interaction may contribute to some of the side effects associated with TKIs.

Summary: Sunitinib inhibits collagen-induced procoagulant activity and delays fibrin formation, which is exacerbated by aspirin. Therefore, we urge awareness of the combined antiplatelet effects of TKIs with aspirin, as this may lead to an increased risk of bleeding.

Summary: This article reports the impact of the CXCL10/CXCR3 pathway on EGFR-TKI resistance in EGFR-mutant lung cancer. It was found that in the co-culture of EGFR-mutant tumor cells and activated PBMCs treated with EGFR-TKI, CXCL10 increased in the supernatant, activating CXCR3 in tumor cells and leading to phosphorylation of Src and NF-KB subunit p65 as well as the expression of HIF-1a. Furthermore, treatment with CXCL10 siRNA decreased CXCL10 levels in the supernatant, suggesting that CXCL10 acts through autocrine and paracrine pathways. Importantly, the elevated CXCL10/CXCR3 signaling was also observed in a transgenic lung cancer mouse model. Overall, the results indicate that increased CXCL10 levels during early EGFR-TKI treatment stimulate persistent tumor cell signaling and contribute to EGFR-TKI resistance via autocrine and paracrine pathways.

Summary: This review discusses the characteristics and analytical methods for determination of sunitinib malate, emphasizing the need for alternative methods that are cost-effective, require minimal sample preparation, consume less toxic solvents, and leave minimal residue. The literature also reveals a lack of research on potential drug-drug pharmacokinetic interactions.

Summary: The classic NF-kappa B pathway is crucial in immune responses and inflammatory diseases. This study showed that IKK beta ubiquitination on lysine-238 increases during inflammation, with USP16 identified as a key regulator affecting p105 phosphorylation. Elevated expression of USP16 in colon macrophages of IBD patients and conditional knockout of USP16 in myeloid cells resulted in reduced severity of IBD, providing insights for targeted intervention therapy.

Summary: This study elucidated the role of MLPH in GBM radiation resistance, showing that MLPH promotes radiotherapy resistance by regulating the NF-kappa B pathway. Additionally, O-GlcNAcylation of MLPH protects it from degradation and stabilizes its presence in cells. These findings provide insights into a potential mechanism of GBM radiation resistance and suggest a therapeutic strategy for GBM treatment.

Summary: In this study, the researchers evaluated the effects of an NF-kappa B inducing kinase (NIK) inhibitor, CW15337, on primary chronic lymphocytic leukemia (CLL) cells, CLL and multiple myeloma (MM) cell lines, and normal B- and T-lymphocytes. They found that CW15337 induced apoptosis and inhibited the non-canonical NF-kappa B signaling pathway, reversing BCL2 family-mediated resistance. The combination of CW15337 with fludarabine or ABT-199 showed synergistic cytotoxic effects.

Summary: The study found that patients with spinal epidural abscess (SEA) were more frail compared to those with back pain, with frailty being determined as an independent risk factor for SEA. The use of the modified frailty index (mFI) may be potentially useful in predicting, prognosticating, and guiding treatment for SEA.
