Characterization of T-Dependent and T-Independent B Cell Responses to a Virus-like Particle

Natural pathogens, such as viruses, often induce T-dependent and T-independent Ab responses. However, the activation and differentiation of Ag-specific B cells under these conditions had not been examined in detail. In this study, we used bacterial phage Q beta-derived virus-like particles (Q beta-VLPs) as an immunogen to examine the T-independent and T-dependent phases of the response in mice. Using Q beta-specific cell labeling and enrichment methods developed in this study, we were able to characterize the rare Ag-specific B cells in detail. Surprisingly, we found that Q beta-VLPs could induce Bcl-6 expression in pregerminal center B cells independently of T cell help. In addition, Q beta-VLP-induced T-independent responses could lead to isotype-switched and somatically mutated memory B cells. Finally, in contrast to what has been reported with several other Ags, long-lived IgG(+) memory cells were induced by Q beta-VLPs, with IgM(+) memory B cells being produced but only evident for a limited time, suggesting that different types of immunogens may preferentially generate or maintain IgM(+) versus IgG(+) memory B cells.

Optogenetic Control of Non-Apoptotic Cell Death

Summary: LiPOP is a set of optogenetic tools that can induce photoswitchable necroptosis and pyroptosis in live cells using light. Studies have shown that combining LiPOPs with upconversion nanoparticles or bioluminescence can achieve wireless optogenetic or chemo-optogenetic killing of cancer cells in multiple mouse tumor models. LiPOPs have the potential to trigger necroptotic and pyroptotic cell death in cultured cells and living animals, opening up possibilities for studying non-apoptotic cell death pathways in microbial infection and anti-tumor immunity.

Connexin32 ameliorates epithelial-to-mesenchymal-transition in diabetic renal tubular via inhibiting NOX4

Metasurface-Based Fiber-to-Chip Multiplexing Coupler

Summary: In this paper, a metasurface-based fiber-to-chip multiplexing coupler is proposed, which enables flexible mode conversion and multiplexing coupling between few-mode fibers and on-chip single-mode waveguides. The approach utilizes the symmetrical characteristics of fiber eigenmodes and the freedom of light phase manipulation to achieve the desired phase distribution of metasurfaces. The use of cylindrical nanopillar as metasurface nanostructures ensures polarization independent optical response. Various fiber-to-chip multiplexing couplers are demonstrated using angular spectrum methods and finite-difference time-domain (FDTD) simulations. It is observed that two or three modes can be demultiplexed and coupled into the corresponding on-chip waveguides with low crosstalk. Furthermore, two possible fabrication schemes for multiplexing couplers are proposed, and the related fabrication tolerances and misalignment effects are evaluated and discussed. Metasurface-based couplers offer advantages of ultracompact footprint and low crosstalk compared to traditional on-chip couplers. This study explores the application of metasurfaces in multiplexing coupling between few-mode fiber and on-chip single-mode waveguides, which is expected to overcome the bottleneck of current mode-division multiplexing technologies in optical interconnection and meet the increasing demand for large data throughput of photonic integrated chips.

HPV Enhances HNSCC Chemosensitization by Inhibiting SERPINB3 Expression to Disrupt the Fanconi Anemia Pathway

Using pH-responsive nanoparticles to deliver SERPINB3 small interfering RNA in combination with cisplatin successfully reverses cisplatin chemotherapeutic resistance.
