The protective effect in rainbow trout (Oncorhynchus mykiss) of an experimental subunit vaccine targeting antigens in the parasite Ichthyophthirius multifiliis has been evaluated and compared to effects elicited by a classical parasite homogenate vaccine. Three recombinant parasite proteins (two produced in E.coli and one in insect cells) were combined and injected i.p., and subsequently, protection and antibody responses were analysed. Both the experimental and the benchmark vaccine induced partial but significant protection against I.multifiliis when compared to control fish. Specific antibody responses of vaccinated trout (subunit vaccine) were raised against one neurohypophysial n-terminal domain protein #10 of three recombinant proteins, whereas the benchmark vaccine group showed specific antibody production against all three recombinant proteins. The immunogenic parasite protein #10 may be a potential vaccine candidate supplementing the protective I-antigen in future vaccine trials.

Summary: Fish parasitological research associated with fisheries and aquaculture has significantly expanded over the past century. The application of parasites as biological tags has provided new insights into fish migration and stock assessments globally. Recent findings include the extension of parasite tags to hosts other than bony fishes, improvements in parasite tag selection, recognition of host traits affecting their use, and the need for integrative multidisciplinary studies combining parasites with classical and modern techniques.

A cVLP-Based Vaccine Displaying Full-Length PCSK9 Elicits a Higher Reduction in Plasma PCSK9 Than Similar Peptide-Based cVLP Vaccines

Summary: Administration of PCSK9-specific monoclonal antibodies and peptide-based PCSK9 vaccines can reduce plasma LDL cholesterol by blocking PCSK9. However, these treatments also increase plasma PCSK9 levels due to the formation of immune complexes. A study using a cVLP-based vaccine platform shows that the FL PCSK9 cVLP vaccine has an enhanced ability to opsonize and clear plasma PCSK9. These findings have implications for the design of vaccines that can effectively mediate opsonization and immune clearance of target antigens.

Summary: The Baltic Sea, a young and ecologically unstable marine ecosystem, exhibits the influence of evolutionary and ecological processes on host-parasite interactions. The increased population of Baltic grey seals has led to higher infection rates of Baltic cod with anisakid nematode larvae. These parasites can be transmitted to humans and pose a risk to human health.

Summary: This study aimed to develop and test a recombinant alternative vaccine based on virus-like particles (VLPs) decorated with VapA protein as the key structural surface protein for combating A. salmonicida bacterial disease. The results showed that rainbow trout vaccinated with VapA-VLP vaccines exhibited comparable protection to the bacterin-based vaccine and produced a strong VapA-specific antibody response.

Summary: Flavobacterium psychrophilum is the causative agent of Bacterial Cold Water Disease (BCWD)/ Rainbow Trout Fry Syndrome (RTFS) in rainbow trout. The disease causes significant mortality in trout farms and leads to economic losses in the aquaculture industry. The immature development of adaptive immunity in young life cycle stages hinders successful vaccination, resulting in the widespread use of antibiotics. Selective breeding and identification of markers associated with disease resistance may improve breeding efforts for more resilient strains.

Summary: This study investigated the functional antibody response and antibody boosting during PM episodes in Aotus monkeys immunized with placental malaria vaccine (PMV) candidates. The results showed that PMV candidates induced functional antibodies with limited heterologous activity in the monkeys, similar to human responses. The Aotus model appears to be suitable for preclinical downselection of PMV candidates and assessment of antibody boosting.

Acute immune responses in zebrafish and evasive behavior of a parasite - who is winning?

Summary: This study investigated the early immune responses in zebrafish larvae infected with the parasite Ichthyophthirius multifiliis. The results showed that the larvae exhibited mild inflammation upon infection, and the 12-day-old larvae were less susceptible compared to the 5-day-old larvae. Neutrophils and macrophages were attracted to the parasites and exhibited neutrophil extracellular traps (NETs) during the fight against the parasite. The parasite employed rotation as an immune evasive strategy, but occasionally, neutrophils and macrophages were able to kill the parasite. In conclusion, the zebrafish larvae responded with localized high activity from neutrophils and macrophages, but only displayed mild inflammation systemically.

Summary: In this study, the glycosylation profiles of anti-S IgG1 Fc in response to mRNA, adenoviral, and protein-based COVID-19 vaccines were compared using mass spectrometry (MS). The results showed variations in the glycosylation patterns between different vaccine platforms and individuals, but overall, vaccine-induced anti-S IgG glycosylation was dynamic with marked overlaps.

Summary: This study found that brass and its pure constituents (copper and zinc) have in vivo and in vitro antiparasitic effects on freshwater parasites. The results showed that brass and pure copper had a 100% mortality rate on parasites within 15 minutes, while zinc had a 100% mortality rate within 30 minutes. The study also found that direct contact with the metal sheets increased the parasiticidal effect. However, further research is needed to investigate the environmental and ecological effects of metal sheet use.

Validation of a QTL associated with resistance to Vibrio anguillarum in rainbow trout (Oncorhynchus mykiss)

Summary: This study demonstrates the feasibility of marker assisted selective breeding of fish resistant to vibriosis, a bacterial disease in rainbow trout farming. A specific single nucleotide polymorphism marker (SNP AX-89945921) associated with resistance was validated. By selecting male fish carrying this marker and breeding them with outbred female trout, a population with higher resistance to the disease was obtained. The QTL fish showed slower and lower morbidity compared to the non-QTL fish when exposed to the bacterium.

Summary: The lifecycle of Thaparocleidus vistulensis, a parasite of European catfish, was studied through observation of infection dynamics, egg development, hatching rates, and survival rates at different life stages. The data obtained allows for predictions of parasite-host dynamics and may contribute to the control of gill-disease in cultured European catfish stocks.
