The plant epidermis or cuticle is constantly exposed to external and internal environmental factors, including an enriched and diverse community of bacteria, yeast, fungi, viruses, and mites. It is not only where the plant has its first physical barrier, but also where organisms can be recognized and potentially where the plant defense responses can be triggered. The plant cuticle is a polymeric composite formed by an array of structurally and chemically heterogeneous compounds, including cutin and wax. A few studies have shown that cuticular components are essential and important drivers of the structure and size of the bacterial community. On the other hand, cuticular components are also important for both pathogens and plants, to initiate the pre-invasion and infection process and to activate the innate immune response, respectively. In this review, we explore current knowledge on the role of the cuticle during the intimate interactions between plants and microorganisms, in particular pathogenic and non-pathogenic bacteria and fungi. Finally, we propose new perspectives on the potential use of this information for agriculture.

Summary: The genus Burkholderia sensu lato consists of a diverse group of bacterial species with unique characteristics such as the presence of two forms of phosphatidylethanolamine (PE) and ornithine lipids (OLs). Only Burkholderia sensu stricto strains constitutively form OLs, while other strains in the group form two forms of PE but no OLs. The absence of OLs in B. cenocepacia affects bacterial growth under specific abiotic stress conditions, while OLs may play an important role in activating the innate immune response in eukaryotes.

Summary: Rare-earth element gadolinium serves as a biostimulant that induces plant defense responses against Botrytis cinerea, modifying both early and late defense responses. The defense induced by gadolinium is more effective compared to calcium-induced response, highlighting the potential of gadolinium as a biocontrol agent against fungal pathogens like B. cinerea.

Summary: This study analyzed the subcellular localization and potential interactors of the Arabidopsis thaliana glycine-rich domain protein 2 (AtGRDP2). It was found that the AtGRDP2 protein is localized in the cytosol and nucleus, and the DUF1399 or RRM domains are sufficient for nuclear localization. Furthermore, heterodimeric interactions between AtGRDP2 and specific proteins were observed in different compartments, suggesting the involvement of AtGRDP2 in post-transcriptional processes.

Arabidopsis thaliana Cuticle Composition Contributes to Differential Defense Response to Botrytis cinerea

Summary: This study revealed that mutants altered in cuticle components exhibit resistance to the pathogen Botrytis cinerea, with cutin mutants showing resistance while wax mutants did not. Transcriptomic analysis showed that genes related to cell wall integrity and ROS accumulation were upregulated in cutin mutants but not in wax mutants. These findings suggest that different defense pathways may be activated in mutants with a resistant phenotype.

Summary: ROP3 has been identified as a positive regulator of infection thread and nodule formation in Lotus japonicus, while not affecting AMF root colonization. The expression of CSSP genes was not impacted by ROP3 in the NFS and AMS conditions, but some rhizobial-specific pathway genes showed decreased expression in the NFS.

Summary: This study isolated and characterized eight Acinetobacter bacteria from the skin of two tropical frog species. It was found that seven isolates had the ability to inhibit the growth of Batrachochytrium dendrobatidis (Bd), while all eight isolates inhibited the growth of the phytopathogen fungus Botrytis cinerea. The study suggests that the frog skin microbiome contains new species of Acinetobacter bacteria with broad antifungal functions.

Summary: Cell wall integrity is crucial for plant defense against biotic and abiotic stresses. Expansins, which modify cell walls, can enhance or weaken plant resistance to pathogens. Changes in cell wall mechanical properties can directly influence the outcome of pathogen infection, while expansins also play a role in triggering defense responses.

Summary: Expression of the TaEPL1 gene in Arabidopsis plants confers increased resistance to bacterial pathogen Pseudomonas syringae and necrotrophic fungus Botrytis cinerea. The expression lines showed reduced disease symptoms and bacterial titers, as well as higher levels of SA- and JA-response genes. EPL1 can be used as an elicitor to generate crops with improved resistance to broad-spectrum diseases.
